- Neotame: Synthesis, stereochemistry and sweetness

Neotame, an alkylated dipeptide-consisting of L,L-stereochemistry, is a new high intensity sweetener being developed by the Nutrition and Consumer Products Sector of Monsanto. The three diastereoisomers of neotame, L,D; D,L; and D,D were synthesized and evaluated for sweetness by sensory testing. Only the L,L-isomer was found to be sweet. The three-dimensional structure of the L,L diastereoisomer determined from single-crystal x-ray analysis demonstrates that the compound exists in an 'L shaped' conformation, previously proposed for the requirement of the sweetness ability by aspartyl- based dipeptide compounds. The confirmation of neotame in the solid state also demonstrates well-oriented hydrophobic and hydrophilic regions which likely account for the high sweetness intensity of this dipeptide compound.

The invention provides a novel compound sodium 3,3-dimethyl-hydroxysulfonate, and a preparation method and application thereof. The molecular formula of the sodium 3,3-dimethyl-hydroxysulfonate is C6H13NaO4S, the molecular weight is 204.2, and the structural formula is disclosed in the specification. The sodium 3,3-dimethyl-hydroxysulfonate can be used as an intermediate for synthesizing neotame and used for preparing neotame. The 3,3-dimethyl-hydroxysulfonate exists in the state of solid double salts, has stabler properties, and is more convenient for storage and transportation.

A crystallization process for forming a stable crystal of N-(3,3-dimethylbutyl)-APM, which comprises using either water or a mixture of water with a lower alcohol as the crystallizing solvent and controlling the crystallization point to 30° C. or above; and another crystallization process therefor, which comprises using either water or a mixture of water with a lower alcohol as the crystallizing solvent and using as the seed crystal a crystal of N-(3,3-dimethylbutyl)-APM exhibiting peculiar peaks of diffracted X-ray at least at diffraction angles (2θ, CuKα ray) of 6.0°, 24.8°, 8.2° and 16.5° to thereby crystallize the above objective crystal preferentially. These crystallization processes enable the constant formation of stable crystals of N-(3,3-dimethylbutyl)-APM at a low cost.

- Process for purification or recovery of sweetener

There is provided a process for purification of N-(3,3-dimethylbutyl)-α-L-aspartyl-L-phenylalanine methyl ester comprising the step of contacting impure N-(3,3-dimethylbutyl)-α-L-aspartyl-L-phenylalanine methyl ester containing at least α-L-aspartyl-L-phenylalanine methyl ester as an impurity, with the mixed solvents having two layers of one layer given from at least one organic solvent which does not mix with water homogeneously and one layer of water to extract N-(3,3-dimethylbutyl)-α-L-aspartyl-L-phenylalanine methyl ester in the organic solvent layer. In the present invention, the desired compound can be purified or recovered efficiently in a high yield without using burdensome steps or operations.
