Green cascade approaches, which utilize sustainable and recyclable heterogeneous biocatalysts, can be used in the catalysis of multicomponent organic reactions to synthesize biologically important substances. Three magnetic nanoparticles, iron (II, III) oxide, cobalt ferrite, and nickel ferrite, were prepared by co-precipitation and functionalized with fibrous silica (KCC-1), and characterized by scanning electron microscopy (SEM), X-ray diffraction (XRD), and vibrating sample magnetometry (VSM). Laccase immobilized on CoFe2O4-KCC-1 was used as a heterogeneous biocatalyst for green and one-pot cascade synthesis of 2-amino-5-oxo-4-aryl-4H,5H-pyrano[3,2-c]chromene-3-carbonitriles. The reaction was performed in the presence of 2,2,6,6-tetramethylpiperidine-1-oxyl (TEMPO) as a synthetic redox mediator. The optimum reaction conditions were found to be as immobilized laccase (100 mg, 95 U) and TEMPO (2 mol-%) in a 100 mm sodium citrate buffer (pH 4.5), 40 °C, and incubation time 17 h. About 80 % of initial activity of the immobilized laccase was retained after 15 independent runs.

The reaction of triethyl(sulfonyl)ammonium chlorosulfonate with 1,1,3,3-tetramethylguanidine (TMG) gave a low-melting complex salt with amphipathic and acidic characters. The salt has a great tendency towards solvation of organic substrates upon slight heating, so it was employed as a homogeneous acidic and functional ionic liquid catalyst in the expedient synthesis of benzo[b]pyrans, pyrano[4,3-b]pyrans, pyrano[3,2-c]quinolones, and pyrido[2,3-d]pyrimidines. The catalytic activity of the complex salt is much better than the simple salts composing it. Spectroscopic studies and chemical analysis of the salt revealed that its empirical formula is in agreement with neutralization of TMG by two H2SO4 molecules. The HSO4 ? anions of the salt seem to form extensive H-bonding with the constituting ammonium cations and Cl?, resulting in melting point depression of the complex salt due to the melting points of pure triethylamine and TMG hydrochloride salts. Another significant feature of this solid salt is that it can preserve its composition even after several times of separation from aqueous extracts and can be handled easily without danger.

After preparation, the promoting ability of this catalyst for one-pot multicomponent synthesis of 2-amino-4H-chromenes, dihydropyrano[3,2-c]chromenes and dihydropyrano[2,3-c]pyrazoles using in situ-developed arylidenemalononitriles through Knoevenagel reaction under solvent-free conditions is described. The sustainable procedure, short reaction times (6–36?min), effortless work-up, non-chromatographic purification, good-to-excellent product yields (79–96%), ease of recovery and reusability of the catalyst up to six reaction runs without a decrease of activity are the notable features of the present protocol.

The catalytic activity of the synthesized catalyst has been explored in the synthesis of a series of biologically and pharmaceutically active pyran-annulated heterocyclic compounds from a one-pot three-component reaction of an aromatic aldehyde, malononitrile/ethyl cyanoacetate, and a C–H activated acidic compound, in the presence of a catalytic amount (10?wt%) of [SBA-16-G]. The catalyst is metal-free, easy to synthesize and to isolate from the reaction mixture, and recycled up to four times without significant loss of catalytic activity. The widespread biological properties of dihydropyrano[3,2-c]chromenes, 2-amino-4-aryl-4H-benzo[h]chromenes, and 3-amino-1-aryl-1H-benzo[f]chromenes have led to increasing efforts for the development of tremendously effective synthetic protocols aimed at their synthesis. In this contribution, sodium malonate was employed for the first time as an efficient catalyst for the one-pot, three-component tandem Knoevenagel–cyclocondensation reaction of 4-hydroxycoumarin (or naphthols), malononitrile/ethyl cyanoacetate, and various aldehydes. These compounds underwent Knoevenagel–Michael–Thorpe–Ziegler cyclization upon heating at 70?°C in water to give the respective medicinally relevant dihydropyrano[3,2-c]chromenes and amino-benzochromenes. The method is versatile and amenable to many substrates as it requires no specialized devices such as microwave, ultrasound and ball-milling. Also, the salient features of this high-yielding protocol are green reaction conditions, the use of commercially available catalyst, easy purification processes by a simple filtration, and relatively shorter reaction times.

Background: Recently, pyrimidine and chromene derivatives have concerned increasing attention because of the wide range of biological activities. Considering the prominence of pyrimidine and chromene derivatives, several procedures have been reported for the synthesis of these compounds. Herein, new magnetic nanoparticles supported ionic liquid has been evaluated for the synthesis of pyrimidine and chromene derivatives. Methods: In this method, pyrimidine and chromene compounds were synthesized by the multi-component reaction of aromatic aldehydes, malononitrile, and active methylene compounds in the presence of the new magnetic nanoparticles supported ionic liquid. The new nanocatalyst was characterized by using TEM, VSM, XRD and IR spectrum. Results: The remarkable advantages of this new procedure involve high yields, short experimental time, low cast and easy preparation of the catalyst and no need of any workup and purification. Magnetic properties of the catalyst led to the easy separation from the reaction mixture by using an external magnet. The catalyst can be reused several times in the desired reaction without significant decrease of its activity. Conclusion: An efficient and green method for the synthesis of chromene and pyrimidine compounds in the presence of magnetic nanoparticles supported ionic liquid have been developed. Thermal stability, low cost and easy synthesis of the catalyst, high yields, easy separation and purification of the product, short experimental time and using a green solvent such as water provided a useful and alternative method for such reactions.

Based on the procedure, it was feasible to synthesize 2-amino-3-cyano-pyrano[3,2-c]chromen-5(4H)-one (4a-4y), 2-amino-3-cyano-pyrano[3,2-c]quinolin-5(4H)-one (6a-6s), 2-amino-3-cyano-7,8-dihydro-4H-chromen-5(6H)-one (8a-8u), 2-amino-3-cyano-pyrano[4,3-b]pyran-5(4H)-one (12a-12f), 2-amino-3-cyano-pyrano[3,2-c]pyridine-6(5H)-one (13a-13f), and 1H-pyrano[2,3-d]pyrimidine-2,4(3H,5H)-dione (14a-14f). The structure of the nanocatalyst was confirmed by various techniques such as IR, SEM, TGA-DTG, EDX, ICP and XRD analyses. In comparison to the conventional methods, the salient features of the present protocol are green reaction conditions, high quantitative yields, short reaction time, high atom economy, low cost, easy isolation of products, and no column chromatographic separation.
