Triazolophanes with benzophenone and S(-)-BINOL functionalization at the intra annular position have been synthesized by the click reaction of 4,4′-bis(azidomethyl) benzophenone with the corresponding bispropargyloxy compounds. The newly synthesized triazolophanes were characterized by spectral and analytical methods. Triazolophane with S(-)-BINOL and benzophenone functionality at the intra annular position shows good target binding ability in molecular docking studies and also better antibacterial activity against all the four tested pathogens viz. Staphylococcus aureus, Bacillus subtilis, Salmonella typhimurium, and Escherichia coli bacteria.

Two new dodecaferrocenyl dendrimers have been prepared using a sixfold Sonogashira coupling reaction and sixfold Huisgen cycloaddition, respectively. In addition, a dendron containing four ferrocenyl groups has been synthesised by conventional synthetic methods. Electrochemical studies have shown that all the ferrocene units are electrochemically equivalent. Moreover, when treated with acid these compounds form multicharged methylium near-infrared-absorbing dendritic dyes with a high extinction coefficient.

- Self-assembled benzophenone bis-urea macrocycles facilitate selective oxidations by singlet oxygen

This manuscript investigates how incorporation of benzophenone, a well-known triplet sensitizer, within a bis-urea macrocycle, which self-assembles into a columnar host, influences its photophysical properties and affects the reactivity of bound guest molecules. We further report the generation of a remarkably stable organic radical. As expected, UV irradiation of the host suspended in oxygenated solvents efficiently generates singlet oxygen similar to the parent benzophenone. In addition, this host can bind guests such as 2-methyl-2-butene and cumene to form stable solid host-guest complexes. Subsequent UV irradiation of these complexes facilitated the selective oxidation of 2-methyl-2-butene into the allylic alcohol, 3-methyl-2-buten-1-ol, at 90% selectivity as well as the selective reaction of cumene to the tertiary alcohol, α,α′-dimethyl benzyl alcohol, at 63% selectivity. However, these products usually arise through radical pathways and are not observed in the presence of benzophenone in solution. In contrast, typical reactions with benzophenone result in the formation of the reactive singlet oxygen that reacts with alkenes to form endoperoxides, diooxetanes, or hydroperoxides, which are not observed in our system. Our results suggest that the confinement, the formation of a stable radical species, and the singlet oxygen photoproduction are responsible for the selective oxidation processes. A greater understanding of the mechanism of this selective oxidation could lead to development of greener oxidants.

- Synthesis and structure-activity analysis of new phosphonium salts with potent activity against African trypanosomes

A series of 73 bisphosphonium salts and 10 monophosphonium salt derivatives were synthesized and tested in vitro against several wild type and resistant lines of Trypanosoma brucei (T. b. rhodesiense STIB900, T. b. brucei strain 427, TbAT1-KO, and TbB48). More than half of the compounds tested showed a submicromolar EC50 against these parasites. The compounds did not display any cross-resistance to existing diamidine therapies, such as pentamidine. In most cases, the compounds displayed a good selectivity index versus human cell lines. None of the known T. b. brucei drug transporters were required for trypanocidal activity, although some of the bisphosphonium compounds inhibited the low affinity pentamidine transporter. It was found that phosphonium drugs act slowly to clear a trypanosome population but that only a short exposure time is needed for irreversible damage to the cells. A comparative molecular field analysis model (CoMFA) was generated to gain insights into the SAR of this class of compounds, identifying key features for trypanocidal activity.

A series of novel linear aromatic ether ketone polymers containing triazole units were synthesized by click chemistry and their structures and properties were characterized by FT-IR, 1H NMR, GPC, TGA, DSC and WAXD. The results showed that these polymers exhibited relatively small molecular weights distributions, good thermal stability and solvent-repelling which could have potential applications as engineering thermoplastic.

- Bisguanidine, Bis(2-aminoimidazoline), and Polyamine Derivatives as Potent and Selective Chemotherapeutic Agents against Trypanosoma brucei rhodesiense. Synthesis and in Vitro Evaluation

The in vitro screening for trypanocidal activity against Trypanosoma brucei rhodesiense of an in-house library of 62 compounds [i.e. alkane, diphenyl, and azaalkane bisguanidines and bis-(2-aminoimidazolines)], which were chosen for their structural similarity to the trypanocidal agents synthalin (1,10-decanediguanidine) and 4,4′-diguanidinodiphenylmethane and the polyamine N1-(3-amino-propyl)propane-1,3-diamine, respectively, is reported. The original synthetic procedure for the preparation of 21 of these compounds is also reported. Most compounds displayed low micromolar antitrypanosomal activity, with five of them presenting a nanomolar inhibitory action on the parasite: 1,9-nonanediguanidine (1c), 1,12-dodecanediguanidine (1d), 4,4′ -bis[1,3-bis(tert-butoxycarbonyl)-2-imidazolidinylimino]diphenylamine (28a), 4,4′-bis(4,5-dihydro-1H-2-imidazolylamino)diphenylamine (28b), and 4,4′-diguanidino-diphenylamine (32b). Those molecules that showed an excellent in vitro activity as well as high selectivity for the parasite [e.g. 1c (IC50 = 49 nM; SI > 5294), 28b (IC50 = 69 nM; SI = 3072), 32b (IC50 = 22 nM; SI = 29.5), 41b (IC50 = 118 nM; SI = 881)] represent new antitrypanosomal lead compounds.

The photophysics of two trichromophoric molecules have been studied by a combination of absorption, fluorescence, phosphorescence, and laser flash photolysis. TRI-1 consists of phenanthrene and naphthalene terminal chromophores joined to a central biphenyl group by methyl ester bridges. Intramolecular singlet-singlet energy transfer (SSET) between the biphenyl and terminal chromophores occurs efficiently with kSSET > 6 ×1010 s-1. Longer range SSET from the naphthalene moiety to the phenanthrene group takes place with a lower rate, k ~ 2.5 × 108 s-1. In TRI-2, the biphenyl moiety is replaced by a benzophenone group. SSET here occurs from the phenanthrene chromophore to the central benzophenone chromophore although with a significantly lower rate than the biphenyl → phenanthrene rate in TRI-1. This is likely due to the change in configuration (ππ* to nπ*) of the excited singlet states involved in the energy transfer. Triplet-triplet (TTET) energy transfer between biphenyl and the terminal chromophores is not observed as a result of a low biphenyl triplet population. That both the phenanthrene and naphthalene triplets are observed following laser photolysis and have different lifetimes and different excitation wavelength concentration ratios indicates that there is no significant TTET between these two groups on the time scale at which the triplets are decaying. This is attributed to the large interchromophore distance (~ 2.5 A) as indicated by modeling studies. In TRI-2, TTET from benzophenone to the terminal chromophores is indicated by both phosphorescence and laser flash photolysis results. Two-laser flash photolysis of the phenanthrene triplet in TRI-1 results in the production of the naphthalene triplet by the following suggested route: (i) production of the upper excited triplet state of the phenanthrene group, (ii) energy transfer to the central biphenyl moiety, and (iii) further energy transfer to the naphthalene chromophore. The utility of this two-laser behavior as the basis for the operation of an optically coupled molecular scale shift register is discussed.
