A siloxane oligomer bearing a chiral crown with high affinity for α-amino acids has been synthesised. The host is a modified form of a coronand first prepared by Cram, in which two 1,1'-binaphthol systems are linked through the oxygen atoms at the 2,2'-positions to form a 22-membered ring system containing six ether oxygen atoms attached to each other by four ethylene units. This was selectively mono-alkenylated to form an undec-10-en-1-yl derivative, which was bonded to linear siloxanes [HSiMe2O(SiMe2O)(n)SiMe2H] and [Me3SiO(Me2SiO)(x)(MeHSiO)(y)SiMe3] with total chain lengths of ca 4 and 200 Si atoms respectively, via a Pt catalysed hydrosilylation reaction.

Molecular editing with fluorine is a validated strategy for modulating the structure and function of organic systems. In the current arsenal of catalytic dihalogenation technologies, the direct generation of the vicinal difluoride moiety from simple olefins without a prefunctionalization step remains conspicuously absent. Herein we report a catalytic, vicinal difluorination of olefins displaying broad functional group tolerance, using inexpensive p-iodotoluene as the catalyst. Preliminary efforts toward the development of an enantioselective variant are also disclosed.

We report the total synthesis of (2S,3R)-2-aminododecan-3-ol has been achieved starting from commercially available 10-undecenoic acid. The key steps involved are Sharpless asymmetric epoxidation, Miyashita's boron-directed C-2 regioselective azidolysis, generated the asymmetric centers and in situ detosylation and reduction of azido tosylate. The antifungal activity of the synthesized (2S,3R)-2-aminododecan-3-ol was evaluated on several Candida strains and was comparable to miconazole, a standard drug.

A new series of liquid crystalline polysiloxanes with a very wide range of mesomorphic temperatures has been prepared. They contain 4-cholesteryl-(10- undecen-1-yloxy) biphenyl-4'-carboxylate mesogenic and 4-biphenyl-4'- allyloxybenzoate mesogenic side groups displaying enantiotropic nematic and chiral nematic phases. The mesomorphic polysiloxane specimen with the widest temperature range was used as the stationary phase in a gas chromatography capillary column and it showed unique separation properties for polycyclic aromatic compounds.

New series of organosiloxane ferroelectric liquid crystalline materials have been synthesized, and their mesomorphic and physical properties have been characterized. These new series contain bis-siloxane or tris-siloxane unit attached to the flexible alkyl chain end of (2S,3S)-2-chloro-3-methylvalerate. The siloxane molecule induction is helpful to the chiral smectic C (S C) formation and chiral SC* stabilization, and it simultaneously causes the liquid crystal temperature range of chiral S C* to be broader. The siloxane member is helpful in reducing the smectic C (SC) transation shift temperature, and the molecule containing tris-siloxane units shows better effect than the bis-siloxane one. The synthesis and characterization of the new FLCs materials which exhibit SC* phase at room temperature and higher spontaneous polarization are presented.

The present invention provides methods for analyzing proteomes, as cells or lysates. The analysis is based on the use of probes that have specificity to the active form of proteins, particularly enzymes and receptors. The probes can be identified in different ways. In accordance with the present invention, a method is provided for generating and screening compound libraries that are used for the identification of lead molecules, and for the parallel identification of their biological targets. By appending specific functionalities and/or groups to one or more binding moieties, the reactive functionalities gain binding affinity and specificity for particular proteins and classes of proteins. Such libraries of candidate compounds, referred to herein as activity-based probes, or ABPs, are used to screen for one or more desired biological activities or target proteins.

- Examples of amphitropic polymers: Monolayer film, Langmuir-Blodgett film and liquid-crystalline properties of some polymeric amphiphiles containing cholestanol moieties and those of some closely related non-polymeric amphiphiles

A range of alternating copolymers were prepared by free-radical-initiated copolymerizations of maleic anhydride with a series of α-alkenes containing cholestanyl moieties. Derivatives of these copolymers were prepared by reacting the anhydride residues with methanol, water, dimethylamine and/or morpholine. A related series of non-polymeric amphiphiles containing steroid moieties was also prepared. Isotherms were measured for monolayers of the various polymers and the various non-polymeric amphiphiles on water and, where possible, Langmuir-Blodgett (LB) multilayers were prepared. The majority of the materials gave good isotherms (relatively steep with collapse pressures >40 mN m-1) indicating that the monolayers were ordered and, as determined by the detection of Bragg peaks by X-ray diffraction, Y-type LB films with regular layer structures. Appropriate materials were also examined, by optical microscopy and differential scanning calorimetry (DSC), for possible liquid-crystalline properties. Four polymers and one non-polymeric amphiphile exhibited smectic A mesophases. Another non-polymeric amphiphile exhibited a cholesteric mesophase. Thus, examples were found of amphitropic polymers and non-polymeric amphiphiles which can form organised molecular arrangements both because they are amphiphilic and because they contain mesogens.

Either (R) or (S) enantiomerically pure methylcarbinol groups are conveniently produced from monosubstituted alkenes via the Sharpless asymmetric dihydroxylation (AD) reaction. The initial AD product, 1,2- dihydroxyalkane, obtained with predictable absolute configuration and high enantiomeric purity, is converted into 2-acetoxy-1-bromoalkane and then subjected to reductive debromination. These conditions are compatible with a variety of functional groups, including acetal, ester, nitrile, ketone, and silyl ether. The advantages of this method are demonstrated by highly efficient, asymmetric syntheses of enantiomerically pure natural products. All four stereoisomers of the WCR sex pheromone 4 are prepared in six steps form nona-1,8-diene in 10-15% overall yield. Similarly, a highly efficient formal total synthesis of antibiotic (-)-A26771B (5) is accomplished via two alternative approaches. The first one transforms dodec-11-enal into enantiomerically pure 5 in 11 steps and 4.1% overall yield, while the second achieves the same transformation in 12 steps and 6.6% overall yield.

Selectively deuterated n-alkyldimethylchlorosilanes were synthesized and used to prepare both chemically modified and physically coated porous silica. 2H NMR spectra were recorded on these modified solids at 46.1 MHz.A sharp liquidlike resonance was observed for the physically coated sample indicative of a complete averaging of the quadrupole interactions.Broad Lorentzian-shaped resonances were observed from all of the chemically modified surfaces with widths less than that expected for rigid-limit conditions.Similar line widths were observed for deuterons in the first and fourth positions and decreases in widths for deuterons toward the free end of the chain.The line shapes are ascribable to the presence of slow motions and the absence of a unique axis of reorientation.Spin-lattice relaxation times also were measured as a function of temperature for the chemically modified surfaces.These latter data as well as line width data indicate that there are at least two different motional domains.
