Despite the increasing need of new antituberculosis drugs, the number of agents approved for the market has fallen to an all-time low. In response to the emerging drug resistance followed, structurally unique chemical entities will be highlighted. decaprenylphosphoryl-β-D-ribose oxidase (DprE1) participating in the biosynthesis of mycobacterium cell wall is a highly vulnerable and validated antituberculosis target. On the basis of it, a systematic strategy was applied to identify a high-quality lead compound (compound 50) that inhibits the essential enzyme DprE1, thus blocking the synthesis of the mycobacterial cell wall to kill M. tuberculosis in vitro and in vivo. Correspondingly, the rational design and synthetic strategy for compound 50 was reported. Notably, the compound 50 has been confirmed to be no toxicity. Altogether, our data suggest the compound 50 targeting DprE1 is a promising candidate for the tuberculosis (TB) therapy.

The invention relates to a synthesis method for organically synthesizing an intermediate N-allylthiourea. The synthesis method mainly comprises: adding 4-5 mol urea, 3 mol allyl alcohol, 12 L of a potassium bromide solution and 1.5 mol alumina powder in a reaction container, controlling the stirring speed at 90-110 rpm, heating the solution to a temperature of 80-90 DEG C, maintaining for 100-130min, carrying out pressure reducing distillation, cooling the solution to a temperature of 10-15 DEG C, precipitating to obtain a crystal, washing with an ether solution, washing with a dimethylaminesolution, re-crystallizing in a methanol solution, and dewatering with a dewatering agent to obtain the finished product N-allylthiourea.

A new procedure for protecting the amide, lactam, urea, and carbamate NH group with a triphenylmethyl (Tr) group is described. The utility of this method is illustrated with molecules that contain other functional groups. A mild deprotection using trifluoroacetic acid makes this a useful method for attaching amide groups on resin for combinatorial synthesis.

The compounds and pharmaceutical compositions of this invention are particularly well suited for inhibiting HIV-1 and HIV-2 protease activity and consequently, may be advantageously used as anti-viral agents against the HIV-1 and HIV-2 viruses. The constant of N-allylthiourea hydrolysis in an alkaline medium at 25 deg C was determined experimentally.Thermodynamic analysis was performed on the conditions for zinc sulfide precipitation by allylthiourea from alkaline solutions of zinc complex salts.The conditions of ZnS precipitation were found experimentally.The structure of the precipitates was determined by means of x-ray phase analysis.
