The N-hydroxyphthalimide derivative-mediated aerobic oxidation of a selection of aldehydes to the corresponding carboxylic acids in air is described. This reaction proceeds via rearrangement of the Creigee (carboxylic peracid) intermediate and/or by the treatment of H2O and/or sulfides. Optimization of reaction conditions established NHNPI (14) as a mild catalyst for the oxidation reaction in MeCN under an atmosphere of air.

- Preparation method of 2-R1 valeric acid

The invention discloses a preparation method for 2-R1 valeric acid. The preparation method comprises the following steps: step 1, with methyl cyanoacetate as a starting material, adding bromopropane and sodium methoxide, carrying out a catalytic reaction, and conducting purifying to obtain 2-cyanomethyl valerate; step 2, subjecting 2-cyanomethyl valerate to a reaction under the catalysis of iodoalkane and sodium methoxide, and conducting aftertreatment to obtain 2-cyano-2-R1 methyl valerate; step 3, enabling the 2-cyano-2-R1 methyl valerate to undergo a reaction in an aqueous solution of sulfuric acid at 120-160 DEG C for 15-40 hours so as to obtain a mixture of 2-R1 valeric acid and 2-R1 methyl valerate; and step 4, hydrolyzing the mixture by using an aqueous sodium hydroxide solution to obtain 2-R1 sodium valerate and methanol, and conducting acidifying by using inorganic acid to obtain 2-R1 valeric acid. Reagents adopted in the preparation method are relatively common, and the risk of the reagents is relatively low; and reaction conditions are mild, and the temperature is easier to control relatively. The invention develops a purification process of the key intermediate 2-cyanomethyl valerate, and the process flow is simple.

- Ruthenium-catalysed hydroxycarbonylation of olefins
-
State-of-the-art catalyst systems for hydroxy- and alkoxycarbonylations of olefins make use of palladium complexes. In this work, we report a complementary ruthenium-catalysed hydroxycarbonylation of olefins applying an inexpensive Ru-precursor (Ru3(CO)12) and PCy3as a ligand. Crucial for the success of this transformation is the use of hexafluoroisopropanol (HFIP) as the solvent in the presence of an acid co-catalyst (PTSA). Overall, moderate to good yields are obtained using aliphatic olefins including the industrially relevant substrate di-isobutene. This atom-efficient catalytic transformation provides straightforward access to various carboxylic acids from unfunctionalized olefins.

- Functionalization of α-C(sp3)?H Bonds in Amides Using Radical Translocating Arylating Groups

α-C?H arylation of N-alkylamides using 2-iodoarylsulfonyl radical translocating arylating (RTA) groups is reported. The method allows the construction of α-quaternary carbon centers in amides. Various mono- and disubstituted RTA-groups are applied to the arylation of primary, secondary, and tertiary α-C(sp3)?H-bonds. These radical transformations proceed in good to excellent yields and the cascades comprise a 1,6-hydrogen atom transfer, followed by a 1,4-aryl migration with subsequent SO2 extrusion.

- Cobalt-Catalyzed Asymmetric Hydrogenation of α,β-Unsaturated Carboxylic Acids by Homolytic H2 Cleavage

The asymmetric hydrogenation of α,β-unsaturated carboxylic acids using readily prepared bis(phosphine) cobalt(0) 1,5-cyclooctadiene precatalysts is described. Di-, tri-, and tetra-substituted acrylic acid derivatives with various substitution patterns as well as dehydro-α-amino acid derivatives were hydrogenated with high yields and enantioselectivities, affording chiral carboxylic acids including Naproxen, (S)-Flurbiprofen, and a d-DOPA precursor. Turnover numbers of up to 200 were routinely obtained. Compatibility with common organic functional groups was observed with the reduced cobalt(0) precatalysts, and protic solvents such as methanol and isopropanol were identified as optimal. A series of bis(phosphine) cobalt(II) bis(pivalate) complexes, which bear structural similarity to state-of-the-art ruthenium(II) catalysts, were synthesized, characterized, and proved catalytically competent. X-band EPR experiments revealed bis(phosphine)cobalt(II) bis(carboxylate)s were generated in catalytic reactions and were identified as catalyst resting states. Isolation and characterization of a cobalt(II)-substrate complex from a stoichiometric reaction suggests that alkene insertion into the cobalt hydride occurred in the presence of free carboxylic acid, producing the same alkane enantiomer as that from the catalytic reaction. Deuterium labeling studies established homolytic H2 (or D2) activation by Co(0) and cis addition of H2 (or D2) across alkene double bonds, reminiscent of rhodium(I) catalysts but distinct from ruthenium(II) and nickel(II) carboxylates that operate by heterolytic H2 cleavage pathways.

Herein, we report a Pd(II)-catalyzed nitrooxylation of unactivated methyl C(sp3)-H bonds using commercial available and easily manageable tert-butyl nitrite as the precursor of ONO2 radical under aerobic conditions. Environmentally benign molecular oxygen is used to initiate the generation of active radical reactant; it is also used as the terminal oxidant. A broad range of nitrooxylated aliphatic carboxamides were prepared in moderate to good yields under mild conditions.

- Carboxylation of benzylic and aliphatic C-H bonds with CO2 induced by light/ketone/nickel

A photoinduced carboxylation reaction of benzylic and aliphatic C-H bonds with CO2 is developed. Toluene derivatives capture gaseous CO2 at the benzylic position to produce phenylacetic acid derivatives when irradiated with UV light in the presence of an aromatic ketone, a nickel complex, and potassium tert-butoxide. Cyclohexane reacts with CO2 to furnish cyclohexanecar-boxylic acid under analogous reaction conditions. The present photoinduced carboxylation reaction provides a direct access from readily available hydrocarbons to the corresponding carboxylic acids with one carbon extension.

- Mild C-H functionalization of alkanes catalyzed by bioinspired copper(ii) cores
-
Three new copper(ii) coordination compounds formulated as [Cu(H1.5bdea)2](hba)·2H2O (1), [Cu2(μ-Hbdea)2(aca)2]·4H2O (2), and [Cu2(μ-Hbdea)2(μ-bdca)]n (3) were generated by aqueous medium self-assembly synthesis from Cu(NO3)2, N-butyldiethanolamine (H2bdea) as a main N,O-chelating building block and different carboxylic acids [4-hydroxybenzoic (Hhba), 9-anthracenecarboxylic (Haca), or 4,4′-biphenyldicarboxylic (H2bdca) acid] as supporting carboxylate ligands. The structures of products range from discrete mono- (1) or dicopper(ii) (2) cores to a 1D coordination polymer (3), and widen a family of copper(ii) coordination compounds derived from H2bdea. The obtained compounds were applied as bioinspired homogeneous catalysts for the mild C-H functionalization of saturated hydrocarbons (cyclic and linear C5-C8 alkanes). Two model catalytic reactions were explored, namely the oxidation of hydrocarbons with H2O2 to a mixture of alcohols and ketones, and the carboxylation of alkanes with CO/S2O82- to carboxylic acids. Both processes proceed under mild conditions with a high efficiency and the effects of different parameters (e.g., reaction time and presence of acid promoter, amount of catalyst and solvent composition, substrate scope and selectivity features) were studied and discussed in detail. In particular, an interesting promoting effect of water was unveiled in the oxidation of cyclohexane that is especially remarkable in the reaction catalyzed by 3, thus allowing a potential use of diluted, in situ generated solutions of hydrogen peroxide. Moreover, the obtained values of product yields (up to 41% based on alkane substrate) are very high when dealing with the C-H functionalization of saturated hydrocarbons and the mild conditions of these catalytic reactions (50-60 °C, H2O/CH3CN medium). This study thus contributes to an important field of alkane functionalization and provides a notable example of new Cu-based catalytic systems that can be easily generated by self-assembly from simple and low-cost chemicals.

- Synthesis of Carboxylic Acids by Palladium-Catalyzed Hydroxycarbonylation

The synthesis of carboxylic acids is of fundamental importance in the chemical industry and the corresponding products find numerous applications for polymers, cosmetics, pharmaceuticals, agrochemicals, and other manufactured chemicals. Although hydroxycarbonylations of olefins have been known for more than 60 years, currently known catalyst systems for this transformation do not fulfill industrial requirements, for example, stability. Presented herein for the first time is an aqueous-phase protocol that allows conversion of various olefins, including sterically hindered and demanding tetra-, tri-, and 1,1-disubstituted systems, as well as terminal alkenes, into the corresponding carboxylic acids in excellent yields. The outstanding stability of the catalyst system (26 recycling runs in 32 days without measurable loss of activity), is showcased in the preparation of an industrially relevant fatty acid. Key-to-success is the use of a built-in-base ligand under acidic aqueous conditions. This catalytic system is expected to provide a basis for new cost-competitive processes for the industrial production of carboxylic acids.

- A biocatalytic method for the chemoselective aerobic oxidation of aldehydes to carboxylic acids

Herein, we present a study on the oxidation of aldehydes to carboxylic acids using three recombinant aldehyde dehydrogenases (ALDHs). The ALDHs were used in purified form with a nicotinamide oxidase (NOx), which recycles the catalytic NAD+ at the expense of dioxygen (air at atmospheric pressure). The reaction was studied also with lyophilised whole cell as well as resting cell biocatalysts for more convenient practical application. The optimised biocatalytic oxidation runs in phosphate buffer at pH 8.5 and at 40 °C. From a set of sixty-one aliphatic, aryl-Aliphatic, benzylic, hetero-Aromatic and bicyclic aldehydes, fifty were converted with elevated yield (up to >99%). The exceptions were a few ortho-substituted benzaldehydes, bicyclic heteroaromatic aldehydes and 2-phenylpropanal. In all cases, the expected carboxylic acid was shown to be the only product (>99% chemoselectivity). Other oxidisable functionalities within the same molecule (e.g. hydroxyl, alkene, and heteroaromatic nitrogen or sulphur atoms) remained untouched. The reaction was scaled for the oxidation of 5-(hydroxymethyl)furfural (2 g), a bio-based starting material, to afford 5-(hydroxymethyl)furoic acid in 61% isolated yield. The new biocatalytic method avoids the use of toxic or unsafe oxidants, strong acids or bases, or undesired solvents. It shows applicability across a wide range of substrates, and retains perfect chemoselectivity. Alternative oxidisable groups were not converted, and other classical side-reactions (e.g. halogenation of unsaturated functionalities, Dakin-Type oxidation) did not occur. In comparison to other established enzymatic methods such as the use of oxidases (where the concomitant oxidation of alcohols and aldehydes is common), ALDHs offer greatly improved selectivity.

- An Efficient Aerobic Oxidation Protocol of Aldehydes to Carboxylic Acids in Water Catalyzed by an Inorganic-Ligand-Supported Copper Catalyst

A method for the aerobic oxidation of aldehydes to carboxylic acids in water by using an inorganic-ligand-supported copper catalyst was developed. This method was performed with the use of atmospheric oxygen as the sole oxidant under extremely mild aqueous conditions, and furthermore, a wide range of aldehydes with various functional groups were tolerated. The copper catalyst could be recycled and used in successive reactions at least six times without any appreciable degradation in performance. This method is operationally simple and avoids the use of high-costing, toxic, air/moisture-sensitive, and commercially unavailable organic ligands. The generality of this method gives it potential to be used on the industrial scale.

- Three-component 1D and 2D metal phosphonates: structural variability, topological analysis and catalytic hydrocarboxylation of alkanes

Herein, we report the use of diphosphonate building blocks and chelating auxiliary N,N-ligands to generate novel polymeric architectures. Specifically, we report new 1D and 2D coordination polymers incorporating three components: transition metal ions (Co2+, Cu2+, Mn2+ or Zn2+), diphosphonate ligands (methane-diphosphonate, MDPA, or 1,2-ethanediphosphonate, EDPA) and N,N-heterocyclic chelators (1,10-phenanthroline, phen, or 2,2′-bipyridine, bpy). Six compounds were isolated under mild synthesis (ambient temperature) conditions: [Cu2(phen)2(EDPA)2(H2O)4]∞ (1), [Co(phen)(EDPA)(H2O)2]∞ (1a), {[Cu(phen)(MDPA)]·H2O]}∞ (2), [Mn(bpy)(EDPA)(H2O)2]∞ (3), [Zn(bpy)(EDPA)]∞ (4), and, lastly, a discrete Ni2+ molecular derivative [Ni(phen)(H2O)4](EDPA) (5). Synthetic details, crystal structures, and intermolecular interactions (π-π stacking and hydrogen bonding) in 1-5 are discussed. Topological analyses and classification of the underlying metal-organic networks in 1-4 were performed, revealing the uninodal 1D chains with the 2C1 topology in 1-3 and the binodal 2D layers with the 3,4L13 topology in 4. In 1-3 and 5, multiple hydrogen bonds lead to the extension of the structures to give 3D H-bonded nets with the seh-4,6-C2/c topology in 1 and 3, 2D H-bonded layers with the 3,5L52 topology in 2, and a 3D H-bonded net with the 6,6T1 topology in 5. The catalytic activity of compounds 1 and 1a was evaluated in a single-step hydrocarboxylation of cyclic and linear C5-C8 alkanes to furnish the carboxylic acids with one more carbon atom. These reactions proceed in the presence of CO, K2S2O8, and H2O at 60 °C in MeCN/H2O medium. Compound 1 showed higher activity than 1a and was studied in detail. Substrate scope was investigated, revealing that cyclohexane and n-pentane are the most reactive among the cyclic and linear C5-C8 alkanes, and resulting in the total yields of carboxylic acids (based on substrate) of up to 43 and 36%, respectively. In the case of cycloalkane substrates, only one cycloalkanecarboxylic acid is produced, whereas a series of isomeric monocarboxylic acids is generated when using linear alkanes; an increased regioselectivity at the C(2) position of the hydrocarbon chain was also observed.

- An Efficient Iron(III)-Catalyzed Aerobic Oxidation of Aldehydes in Water for the Green Preparation of Carboxylic Acids

The first example of a heterogeneous iron(III)-catalyzed aerobic oxidation of aldehydes in water was developed. This method utilizes 1 atmosphere of oxygen as the sole oxidant, proceeds under extremely mild aqueous conditions, and covers a wide range of various functionalized aldehydes. Chromatography is generally not necessary for product purification. Its operational simplicity, gram-scale oxidation, and the ability to successively reuse the catalyst, make this new methodology environmentally benign and cost effective. The generality of this methodology gives it the potential to be used on an industrial scale.

The stereoselective desymmetrisation of achiral 2-alkyl-1,3-diols is performed by oxidation of one of the two enantiotopic primary alcohol moieties by means of Acetobacter aceti MIM 2000/28 to afford the corresponding chiral 2-hydroxymethyl alkanoic acids (up to 94 % ee). The procedure, carried out in aqueous medium under mild conditions of pH, temperature and pressure, contributes to enlarge the portfolio of enzymatic oxidations available to organic chemists for the development of sustainable manufacturing processes.

- Structurally well-characterized new multinuclear Cu(II) and Zn(II) clusters: X-ray crystallography, theoretical studies, and applications in catalysis

Two new trinuclear Cu(ii) and dinuclear Zn(ii) clusters are crystallized out by reacting the metal salts with the triethanolamine (H3tea) ligand in the presence of benzoic acid (Hba). The complexes are characterized by elemental, thermal, magnetic, spectral (FTIR, UV-Visible, EPR, and photoluminescence) and single crystal X-ray studies. Single crystal X-ray crystallography reveals the composition of the complexes to be [Cu3(H2tea)2(ba)2(NO3)2] (1) and [Zn2(H2tea)(ba)3]H2O (2). FTIR ascertains the binding modes of H2tea-, ba- and NO3-. Triethanolamine binds in both the complexes in the monoanionic (H2tea-) mode. ba- is present as an anchoring auxiliary to generate di- and trinuclear clusters. The Cu(ii) ion is present as a distorted octahedral center in the Cu3 cluster (1), while the two Zn(ii) ions in 2 have been reported for the first time to possess distorted octahedral as well as tetrahedral geometry in the same molecule. The intriguing features of the non-covalent supramolecular interactions have been investigated and supported theoretically by using Hirshfeld surface analysis and ab initio methods. The solid state photoluminescence (PL) spectra of 1 and 2 disclose the luminescence property of the complexes. Due to the closed or nearly closed shell configuration (d9 or d10), the present complexes are screened for catalytic properties in the hydrocarboxylation of alkanes and cycloalkanes. The catalytic activity data are indicative of the potential catalytic properties of 1 and 2.

- Systematic methodology for the development of biocatalytic hydrogen-borrowing cascades: Application to the synthesis of chiral α-substituted carboxylic acids from α-substituted α,β-unsaturated aldehydes
-
Ene-reductases (ERs) are flavin dependent enzymes that catalyze the asymmetric reduction of activated carbon-carbon double bonds. In particular, α,β-unsaturated carbonyl compounds (e.g. enals and enones) as well as nitroalkenes are rapidly reduced. Conversely, α,β-unsaturated esters are poorly accepted substrates whereas free carboxylic acids are not converted at all. The only exceptions are α,β-unsaturated diacids, diesters as well as esters bearing an electron-withdrawing group in α- or β-position. Here, we present an alternative approach that has a general applicability for directly obtaining diverse chiral α-substituted carboxylic acids. This approach combines two enzyme classes, namely ERs and aldehyde dehydrogenases (Ald-DHs), in a concurrent reductive-oxidative biocatalytic cascade. This strategy has several advantages as the starting material is an α-substituted α,β-unsaturated aldehyde, a class of compounds extremely reactive for the reduction of the alkene moiety. Furthermore no external hydride source from a sacrificial substrate (e.g. glucose, formate) is required since the hydride for the first reductive step is liberated in the second oxidative step. Such a process is defined as a hydrogen-borrowing cascade. This methodology has wide applicability as it was successfully applied to the synthesis of chiral substituted hydrocinnamic acids, aliphatic acids, heterocycles and even acetylated amino acids with elevated yield, chemo- and stereo-selectivity. A systematic methodology for optimizing the hydrogen-borrowing two-enzyme synthesis of α-chiral substituted carboxylic acids was developed. - How to force a classical chelating ligand to a metal non-chelating bridge: The observation of a rare coordination mode of diethanolamine in the 1D complex {[Cu2(Piv)4(H3tBuDea)](Piv)}n

The novel chain coordination polymer {[Cu2(Piv) 4(H3tBuDea)](Piv)}n (1) has been prepared through the self-assembly reaction of copper(ii) nitrate with pivalic acid (HPiv) and N-tert-butyldiethanolamine (H2tBuDea) in acetonitrile solution. Crystallographic analysis revealed the extremely rare non-chelating bridging coordination mode of diethanolamine ligand in 1, observed for the first time in transition metal complexes, as well as in complexes of diethanolamine having a non-coordinating aliphatic group at the N atom. Possible reasons for such a coordination and analysis of the main coordination modes of diethanolamine-based ligands are discussed. Variableerature (1.8-300 K) magnetic susceptibility measurements showed that 1 represents a rare example of dicopper(ii) tetracarboxylate that is a diamagnetic solid at room temperature. This behaviour is compared with literature examples and discussed on the basis of DFT calculations. Furthermore, 1 acts as an efficient catalyst for the mild hydrocarboxylation of linear and cyclic C5-C8 alkanes into the corresponding carboxylic acids.

A new methodology for the oxidation of aldehydes promoted by commercially available Fe3O4 nanoparticles (Fe3O4 NPs) activated by ethyl acetoacetate was developed. The use of ethyl acetoacetate as additive was crucial to achieve high reactivities. All reactions were realized under solvent free conditions, using air or tBuOOH as oxidants. Finally, the separation and reuse of the magnetically recoverable nanoparticles make this methodology very practical, simple and economical.

- Oxidative degradation of fragrant aldehydes. Autoxidation by molecular oxygen

The oxidative degradation of fragrant aldehydes by molecular oxygen has been investigated. The oxygen consumption was monitored and the bond dissociation energy (BDE) of the aldehyde C(O)-H bond were calculated by DFT method. The oxidation products were identified by GC/MS. The different pathways accounting for the oxidative degradation are discussed. The main product is the acid, beside the formate ester. Both oxidation products result from the Baeyer-Villiger reaction involving a peracid R(CO)OOH whereas minor products arise from the hydroperoxide ROOH intermediate derived either from the acyl peroxy radical, R(CO)OO or from the decarboxylation of the peracid RC(O)OOH.

- Tautomeric effect of hydrazone Schiff bases in tetranuclear Cu(ii) complexes: Magnetism and catalytic activity towards mild hydrocarboxylation of alkanes

Three new tetranuclear copper(ii) complexes [Cu(HL1)] 4·4EtOH (1·4EtOH), [Cu(HL2)]4 (2) and [Cu(H2L3)]4(NO3) 4·2H2O (3·2H2O) have been synthesized using three different hydrazone Schiff base ligands derived from the condensation of the aromatic acid hydrazides 2-hydroxybenzo-, 2-aminobenzo- or benzo-hydrazide, with 2,3-dihydroxybenzaldehyde. Complexes 1 and 3 have been characterized by single crystal X-ray diffraction analysis. The coordinating behaviour of the ligand depends on the nature of the ortho substituent present in the hydrazide moiety. The ligands bearing a strong electron donating group (by resonance) in the ortho position undergo complexation via enolization and deprotonation, whereas the absence of such an effect leads to complexation via the keto form, and two different types of tetranuclear Cu(ii) clusters, viz. open-cubane and cubane, are obtained. Variable temperature magnetic susceptibility measurements of complexes 1 and 3 have been carried out to examine the nature of magnetic interaction between the Cu(ii) centres. All the three complexes (1-3) act as good catalyst precursors towards mild hydrocarboxylation of linear and cyclic alkanes into carboxylic acids in water-acetonitrile medium.

- Cooperative N-Heterocyclic Carbene (NHC) and Ruthenium Redox Catalysis: Oxidative Esterification of Aldehydes with Air as the Terminal Oxidant
-
The paper describes a cooperative NHC (N-heterocyclic carbene) and ruthenium-based redox catalysis for the mild aerobic oxidative esterification of various aromatic and heteroaromatic aldehydes. The ruthenium(II) complex Ru(bpz)3(PF6)2 (bpz=2,2′-bipyrazine) as catalyst is shown to be compatible with free NHCs. The NHC is used in these cascade reactions for the umpolung of the aldehyde to form the corresponding Breslow intermediate which in turn gets oxidized to an acylazolium ion by the ruthenium redox catalyst. Air is used as a terminal oxidant for oxidation (regeneration) of the ruthenium catalyst. In addition, we will show that in the absence of the ruthenium redox catalyst and alcohol, NHC-catalyzed aerobic oxidation of aldehydes delivers the corresponding acids in good to excellent yields. The manganese-containing catalytic system [MnIV,IV 2O3(tmtacn)2]2+ (1)/carboxylic acid (where tmtacn=N,N′,N′′-trimethyl-1,4,7-triazacyclononane), initially identified for the cis-dihydroxylation and epoxidation of alkenes, is applied for a wide range of oxidative transformations, including oxidation of alkanes, alcohols and aldehydes employing H2O2 as oxidant. The substrate classes examined include primary and secondary aliphatic and aromatic alcohols, aldehydes, and alkenes. The emphasis is not primarily on identifying optimum conditions for each individual substrate, but understanding the various factors that affect the reactivity of the Mn-tmtacn catalytic system and to explore which functional groups are oxidised preferentially. This catalytic system, of which the reactivity can be tuned by variation of the carboxylato ligands of the in situ formed [MnIII,III 2(O)(RCO2)2(tmtacn)2]2+ dimers, employs H2O2 in a highly atom efficient manner. The oxidation of alcohols to aldehydes using stoichiometric 4-acetamido-2,2,6,6-tetramethylpiperidine-1-oxoammonium tetrafluoroborate (1) in CH2Cl2 at room temperature is a highly selective process favoring reaction at the carbinol center best able to accommodate a positive charge. The oxidation of aldehydes to carboxylic acids by 1 in wet acetonitrile is also selective; the rate of the process correlates with the concentration of aldehyde hydrate. A convenient and high yield method for oxidation of alcohols directly to carboxylic acids has been developed.

The new two-dimensional (2D) heterometallic CuII/Mg coordination polymer [Cu2Mg2(μ-Htea)2(μ6- pma)(H2O)6]n?6nH2O (1) with an unprecedented [Cu2Mg(μ-O)2(μ-COO)2] - core has been easily generated by aqueous medium self-assembly from copper(II) nitrate, triethanolamine (H3tea), magnesium hydroxide, and pyromellitic acid (H4pma). The crystal structure of 1 is composed of infinite interdigitated 2D metal-organic layers that extend via H-bonds into an intricate 3D supramolecular framework. The topological analysis of 1 discloses a binodal 2,4-connected underlying 2D net with the unique topology described by the point symbol of (64?8?10)(6), further simplification of which leads to an uninodal 4-connected net with the sql topology. Apart from representing a very rare example of the heterometallic Cu/Mg coordination network, compound 1 also acts as an efficient catalyst precursor for the mild single-pot hydrocarboxylation of linear and cyclic C n (n = 5-9) alkanes into the corresponding Cn+1 carboxylic acids.

- Mild oxidative functionalization of alkanes and alcohols catalyzed by new mono- and dicopper(II) aminopolyalcoholates

The new mono- and dicopper(II) complexes [Cu(H3L 1)(NCS)] (1) and [Cu2(μ-HL2) 2(NCS)2] (2) were easily self-assembled from Cu(CH 3COO)2·H2O, NaNCS, NaOH and N,N,N′,N′-tetrakis(2-hydroxyethyl)ethylenediamine (H 4L1) or N-ethyldiethanolamine (H2L 2), respectively. They were fully characterized by IR spectroscopy, ESI-MS(±), elemental and single-crystal X-ray diffraction analyses, and applied as homogeneous catalysts for (i) the oxidation of alkanes by t-BuOOH in air to alkyl peroxides, alcohols and ketones, and in turn the oxidation of alcohols to ketones, and (ii) the single-pot aqueous medium hydrocarboxylation, by CO, H2O and K2S2O8, of various linear and cyclic Cn (n = 5-8) alkanes into the corresponding C n+1 carboxylic acids. Compound 1 was significantly more active in the oxygenation of alkanes and oxidation of alcohols, allowing to achieve 18% yield (TON = 800) of oxygenates in the oxidation of cyclohexane, and 78% yield (TON = 780) of cyclohexanone in the oxidation of cyclohexanol. In alkane hydrocarboxylations, 1 and 2 exhibited comparable activities with the total yields (based on alkane) of carboxylic acids attaining 39%. The selectivity parameters for oxidative transformations were measured and discussed, supporting free-radical mechanisms.

- Mild, single-pot hydrocarboxylation of linear C5-C9 alkanes into branched monocarboxylic C6-C10 acids in copper-catalyzed aqueous systems

A single-pot method has been developed for the hydrocarboxylation of the liquid C5-C9 alkanes (n-pentane, n-hexane, n-heptane, n-octane, n-nonane and 3-methylhexane) into the branched monocarboxylic C 6-C10 acids bearing one more carbon atom. This method is characterized by a direct, selective and low-temperature (60 °C) hydrocarboxylation reaction of the alkane with carbon monoxide, water (which acts as a reagent besides being a solvent component) and potassium peroxodisulfate, in H2O/MeCN medium. The hydrocarboxylations are markedly enhanced in the presence of a tetracopper(II) triethanolaminate complex as a homogeneous catalyst precursor. Total yields (based on alkane) of carboxylic acids up to 46% (with 97-99% overall selectivity) have been achieved, which are remarkable in the field of alkane functionalization under mild conditions, especially for a C-C bond formation reaction in aqueous acid-solvent-free medium. The regio- and bond selectivity parameters have been determined and a free radical mechanism has been proposed.

- Syntheses and evaluation of anticonvulsant profile and teratogenicity of novel amide derivatives of branched aliphatic carboxylic acids with 4-aminobenzensulfonamide
-
Despite the availability of 14 new antiepileptic drugs (AEDs), about 30% of epileptic patients are not seizure-free. Consequently there is substantial need to develop new effective AEDs. A novel class of aromatic amides composed of phenylacetic acid or branched aliphatic carboxylic acids, with five to nine carbons in their carboxylic moiety, and aminobenzenesulfonamide were synthesized and evaluated in the anticonvulsant rat-maximal electroshock (MES) and subcutaneous metrazol seizure (scMet) tests. Fourteen of the synthesized amides had an anticonvulsant ED50 of 50 values of 7.6, 9.9, and 9.4 mg/kg and remarkable protective index (PI = TD 50/ED50) values of 65.7, 50.5, and 53.2, respectively. These potent sulfanylamides caused neural tube defects only at doses markedly exceeding their effective dose. The anticonvulsant properties of these compounds make them potential candidates for further development as new, potent, and safe AEDs.

- Synthesis and evaluation of antiallodynic and anticonvulsant activity of novel amide and urea derivatives of valproic acid analogues
-
Valproic acid (VPA, 1) is a major broad spectrum antiepileptic and central nervous system drug widely used to treat epilepsy, bipolar disorder, and migraine. VPA's clinical use is limited by two severe and lifethreatening side effects, teratogenicity and hepatotoxicity. A number of VPA analogues and their amide, N-methylamide and urea derivatives, were synthesized and evaluated in animal models of neuropathic pain and epilepsy. Among these, two amide and two urea derivatives of 1 showed the highest potency as antineuropathic pain compounds, with ED50 values of 49 and 51 mg/kg for the amides (19 and 20) and 49 and 74 mg/kg for the urea derivatives (29 and 33), respectively. 19, 20, and 29 were equipotent to gabapentin, a leading drug for the treatment of neuropathic pain. These data indicate strong potential for the above-mentioned novel compounds as candidates for future drug development for the treatment of neuropathic pain. 2009 American Chemical Society.

- Structure and reactivity in the hydrolyses of aliphatic carboxylic acid esters and chlorides

For hexanoic acid and its seven isomers, relative rates have been determined for acid catalysed esterification with methanol, and compared with those for saponification of the methyl esters. A good correlation between logarithms of relative rates for the two reactions is obtained, and it is suggested that the eight isomers provide a test set of compounds in which steric effects alone act on reactivity at the acyl carbon. A full set of steric parameters (Es′ values) are presented. Rates of solvolyses of the acid chlorides- of the isomers have been determined conductometrically in 3:1 wt:wt acetonitrile water. Logarithms of relative rates show a poor correlation with Es′, and, taking into account the solvent dependence of the rates, the pattern excludes both rate-limiting formation of a tetrahedral intermediate and rate-limiting dissociation of chloride to form acylium ions. The remaining possibilities, a concerted process (ANDN?) and rapid reversible formation of a hydrate followed by rate-limiting dissociation of chloride (AN + DN?,) are considered. Copyright

The recombinant polyketide synthase thioesterase domains from the pimaricin and 6-deoxyerythronolide B biosynthetic pathways catalyze hydrolysis of a number of simple N-acetylcysteamine thioester derivatives. This study demonstrates that thioesterases are not highly substrate selective in formation of the acyl-enzyme intermediate, in contrast to non-ribosomal peptide synthase thioesterase domains that show very high specificity for substrate loading. This observation has important implications for the engineering of biosynthetic pathways to produce polyketide products.

- Vanadium-catalyzed carboxylation of linear and cyclic C5 and C6 alkanes

Cyclopentane, cyclohexane, pentane, and hexane are carbonylated in single-pot processes and under mild conditions to carboxylic acids (highest yields of 54-33% and turnover numbers [TONs] of 76-50) by vanadium (IV) and (V) complexes in TFA. These complexes present N,O- or O,O-ligands, namely basic forms of aminoalcohols and of (hydroxyimino)dicarboxylic acids, trifluoroacetate, or triflate. The effects of various parameters (e.g., catalyst type, oxidizing agent, CO pressure, temperature, reaction time, type of solvent) were investigated. The use of either too low or too high CO pressures is discouraged, because the former promote the formation of trifluoroacetate esters and the latter, above a certain level, do not result in higher yields or TONs of the carboxylic acids. Carbon- and oxygen-centered radical mechanisms are suggested by experiments with radical traps and by acid product distribution.

Depending on the substrate nitrile 2 and/or amide 3 accompanied with carboxylic acid 4 and parent carbonyl compounds 5 were obtained. Formation of nitriles 2 with H2O2-acetonitrile system is limited for a few more active substrates. The mechanism of the reaction, based on generated in situ peroxyiminoacetic acid, is presented. A broad spectrum of aliphatic, unsaturated and aromatic nitriles 2 was obtained by oxidation of corresponding N,N-dimethylhydrazones 1 with hydrogen peroxide in the presence of poly(bis-9,10-anthracenyl) diselenide (PADS) (7) as catalyst.

Aerobic oxidation of various alcohols has been accomplished by using a new catalytic system. N-hydroxyphthalimide (NHPI) combined with Co(acac)3. The oxidation of alcohols by NHPI was found to be markedly enhanced by adding a slight amount of Co(acac)3 (0.05 equiv. to NHPI). Thus, secondary alcohols and vic-diols which are difficult to be oxidized by NHPI alone were smoothly oxidized with molecular oxygen (1 atm) to the corresponding carbonyl compounds under relatively mild conditions (65 ~ 75 °C).

The invention provides novel compounds having the formula: STR1 wherein R when taken individually is H; R1 when taken individually is H or OH; R and R1 when taken together represent a double bond; R2 is an alpha-branched C3 -C8 alkyl, alkenyl, alkynyl, alkoxyalkyl or alkylthioalkyl group; a C3 -C8 cycloalkyl, C5 -C8 cycloalkenyl or C5 -C8 cycloalkylalkyl group, any of which may be substituted by methylene or one or more C1 -C4 alkyl groups or halo atoms; or a 3 to 6 membered oxygen or sulphur containing heterocyclic ring which may be substituted by one or more C1 -C4 alkyl groups or halo atoms; R3 is hydrogen or methyl; R4 is H or 4'-(alpha-L-oleandrosyl)-alpha-L-oleandrosyloxy with the proviso that when R2 is alkyl it is not isopropyl or sec-butyl; when R4 is H, each of R and R1 is H, and R2 is not methyl or ethyl; and when R4 is H, R is H, R1 is OH, and R2 is not 2-buten-2-yl, 2-penten-2-yl or 4-methyl-2-penten-2-yl. The compounds are broad spectrum antiparasitic agents having utility as anthelmintics, ectoparasiticides, insecticides and acaricides. The invention also provides a process for producing the novel avermectin and milbemycin derivatives by adding a carboxylic acid or derivative thereof to a fermentation of an avermectin or milbemycin producing organism.

For the first time, a highly efficient and direct oxidation of a variety of organoborane intermediates into carboxylic acids has been demonstrated, without an increase or decrease of carbon atoms.Synthetically useful procedures have been developed for the ready conversion of representative terminal alkenes into carboxylic acids via oxidation of the organoboranes obtained by hydroboration of the terminal alkenes.The oxidation works well with a variety of organoboranes derived from reagents such as dibromoborane-methyl sulfide (HBBr2*SMe2), monobromoborane-methyl sulfide (H2BBr*SMe2), monochloroboran-methyl sulfide (H2BCl*SMe2), borane-methyl sulfide (H3B*SMe2), thexylborane (H2BThx), and dicyclohexylborane (HBChx2).The oxidation is achieved in a convenient manner with pyridinium dichromate (PDC), sodium dichromate in aqueous sulfuric acid (Na2Cr2O7-H2SO4) and chromium trioxide in 90 percent aqueous acetic acid (CrO3-HOAc-H2O).These oxidations afford carboxylic acids in very good yields with complete retention of the structure of the organic group attached to boron.
