- Synthetic Method of Eclanamine maleate
Eclanamine maleate (CAS NO.: 71027-14-0), with its systematic name of (+-)-trans-3',4'-Dichloro-N-(2-(dimethylamino)cyclopentyl)propionanilide maleate (1:1), could be produced through many synthetic methods.
Following is one of the reaction routes:
Cyclopentene oxide (I) could react with dimethylamine (II) to produce 2-(dimethylamino)cyclopentanol (III), and the yielding product is treated with methanesulfonyl chloride and NaH in THF to give the corresponding methasulfonyl ester (IV). The condensation of (IV) with 3,4-dichloroaniline (V) in THF yields N,N-dimethyl-N-(3,4-dichlorophenyl)cyclopentane-1,2-diamine (VI), which is acylated with propionic anhydride (A) and treated finally with maleic acid (B).
