Based on these results and the previous literature,[19]
the proposed reaction pathway is illustrated in
Scheme 4. Bases such as Cs2CO3 can generate the
enolate A from the imino ester by abstracting the α-
proton followed by conjugate addition of the enolate
to benzylidenepyrazolone. The resulting enolate
intermediate B can undergo aromatization to produce
intermediate C, which next undergoes protonation to
produce the conjugate adduct (Scheme 4).
In conclusion, a novel method for the chiral
synthesis of unsymmetric β,β-diaryl-α-amino acid
esters via enantioselective conjugate addition of
benzylidenepyrazolones and glycine bases under
Copper(I)/iPr-Phosferrox complex as the catalyst has
been reported. A series of unnatural enantioenriched
β,β-diaryl-α-amino acid esters with two continuous
stereocenters are obtained with good to high yields
and enantioselectivities. More importantly, this
protocol is amenable to gram-scale synthesis of novel
unnatural amino acid esters, and catalyst loading can
be lowered to 1 mol%. The corresponding adducts
can be easily converted to enantiopure vicinal amino
alcohols, showing great synthetic potential.

Under argon atmosphere, the solution of planar-chiral
ferrocene P, N-ligand (10 mol%) (0.01 mmol) and
Cu(CH3CN)4BF4 (0.01 mmol) in 1 mL THF was stirred at

the starting material as monitored by TLC. After removal
of the solvent, the residue was subjected to
chromatography on a silica gel column (60–120 mesh)
using 10:1 petroleum ether-ethyl acetate solvent mixture as
eluent to afford 3.
