Fluorimetric measurements are carried out in situ on the 4-chloro-7-nitrobenzo-2-oxa-1,3-diazole [10199-89-0] derivs. formed after thin-layer chromatog. of the clarified fermn. broth. This fluorimetric procedure is 800-fold as sensitive as the analogous ninhydrin procedure. L-methionine [63-68-3] is shown to be involved in the methylation of gentamicin C1a [26098-04-4] to C2 [25876-11-3] and then to C1 [25876-10-2].

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) The sulfonylurea antidiabetic glibornuride (I) [26944-48-9] was detd. by extn. with isoamyl acetate from serum or plasma, heating for 15 min at 140.degree. with 7-chloro-4-nitrobenzo-2-oxa-1,3-diazole (II) [10199-89-0], and fluorometric detn. of the reaction product (excitation at 466, emission at 510 nm). Since II itself is not fluorescent, an excess of the reagent does not distrub the method. Details are given of the anal. procedures, including effect of the temp. and time of heating, pretreatment of the isoamyl acetate with NaOH, etc.Several substances with their cas registry numbers 10238-21-8 and 26944-48-9 may be metioned in this study. The method allows the detn. of I concns. obtained after therapeutic doses of I. The lower limit of detection was 40 ng/mL, the recovery .apprx.100%. The coeffs. of variation for the detn. of I at 0.1-1 .mu.g/mL ranged 4.1-12.1%. The same method could be used to det. tolbutamide [64-77-7] and glibenclamide [10238-21-8] in plasma or serum. These drugs gave fluorescence values, relative to I, of 110 and 90%, resp. .
