The DA activity was assessed by measurement of the presynaptic metabolite 3,4-dihydroxyphenylacetic acid [102-32-9] using differential pulse voltammetry in free moving prepns. Tail-pinch lasting 5 min or 10 min was followed by increases in DA activity of 25% and 35%, resp. Three types of social interaction were used: interaction with an aggressive male congener, interaction with a nonaggressive male congener, or interaction with a female congener. The greatest increase in dopaminergic activity (40%) was obsd. after the aggressive interaction. The reactivity of the DA neurons within the nucleus accumbens is, therefore, dependent on the characteristics of the behavioral situation.

Seven days after a single intracerebral ventricular injection of 100 mg of I, the concn. of NE in all brain regions examd. was reduced markedly, as was the concn. of DA in the median eminence. The concns. of DA and dihydroxyphenylacetic acid in regions other than the median eminence, and the concns. of 5-hydroxytryptamine and 5-hydroxyindoleacetic acid in all regions analyzed were unaltered. Multiple injections of I (100 mg every 48 h ′ 3) did not increase the NE depletion, but caused slight decreases in DA and 5-hydroxytryptamine in some regions. The NE concns. in hypothalamic nuclei were reduced significantly 4 h after 100 mg administration of I and this depletion was maintained for ~28 days. I-induced decline of NE in all brain regions, but not the decline of DA in the median eminence, was prevented partially by pretreatment with desipramine. I is a relatively specific neurotoxin for NE neurons, being particularly effective in destroying NE nerve terminals in the hypothalamus.
