DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The binding of 6 cephalosporins (cefotaxime [63527-52-6], cefuroxime [55268-75-2], cefazoline [25953-19-9], cephalothin [153-61-7], cephaloridine [50-59-9], and cephacetrile [10206-21-0]) to human serum albumin was studied by fluorescence spectrophotometry. Three binding sites for the drugs were identified by the fluorescence probe technique; however, 4 binding sites were detected by UV spectrophotometry. The binding consts. of cephalosporins to human serum albumin measured by fluorescence probe technique were higher than those measured by UV spectrophotometry.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Epileptogenic activity induced by i.v. injection of certain cephalosporin derivs. was studied. Ceftezole [26973-24-0] provided the most potent epileptogenic activity among the drugs tested. Changes in seizure patterns on EEG tracings and behavioral signs after administrations of cefotiam [61622-34-2] and cefazolin [25953-19-9] were similar to those seen after ceftezole, though the intensity and duration were less than those of ceftezole. Cephacetrile [10206-21-0] and cephaloridine [50-59-9] elicited the spiking activity in the frontal cortex and the other regions without apparent behavioral changes. Latamoxef [64952-97-2] and cefmenoxime [65085-01-0] displayed a weak epileptogenic activity at a dose of 1000 mg/kg. On the other hand, cephapirin [21593-23-7], cefmetazole [56796-20-4] and cefoxitin [35607-66-0] did not evoke any changes in EEG nor in behavior. Penicillin G [61-33-6] at a dose of 200 mg/kg affected spike or spike-and-wave complex in a few cases, but at a dose of 500 mg/kg the animals died of dyspnea almost immediately after injection without showing apparent epileptogenic signs. These results suggest that some of cephalosporins such as ceftezole, cefotiam, cephacetrile, and cefazolin provide epileptogenic activity at higher doses.
