The selective activity of I and II against varicella zoster virus is apparently related to the induction of a pyrimidine deoxyribonucleoside kinase by the virus similar to that induced by herpes simplex virus.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) The halogenated pyrimidine analog 5-bromo-2'-deoxycytidine (I) [1022-79-3] was infused into BALB/C mice bearing EMT-6 tumors via the intravenous or intraarterial route. Hepatic dehalogenation of the I was blocked by 5-diazouracil (II) to ascertain its relative importance in the degrdn. of intravenously administered analogs. Increased radiosensitization was noted with higher intravenous pyrimidine concns., but II blockage of dehalogenation had little effect. Following intravenous infusion, enough I apparently bypasses the hepatic vessels to permit tumor radiosensitization despite diln. of the drug by the systemic circulation.
