DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The effects of repeated treatment of rats with the antidepressant or potential antidepressant agents alaproclate [60719-82-6], citalopram [59729-33-8], clomipramine [303-49-1], desipramine [50-47-5], imipramine [50-49-7], maprotiline [10262-69-8], mianserin [24219-97-4] and zimeldine [56775-88-3] on convulsions released by decapitation were examd. The noradrenaline uptake inhibitors desipramine, imipramine and maprotiline increased significantly the latency of onset of the post-decapitation convulsions (PDC's) after repeated administration of 10 mmol/kg orally twice daily, or 66 mmol/kg orally once daily (desipramine), for 15 days. The duration of the PDC's was slightly prolonged by these agents. A single acute dose of desipramine (20 mg/kg) administered at various time intervals before decapitation (1 to 24 h) had no effect on the PDC's; repeated treatment with alaproclate, citalopram, clomipramine, mianserin and zimeldine did not have any effect upon the PDC latency. The results are interpreted as evidence for noradrenaline receptor subsensitivity following chronic treatment.

DOCUMENT TYPE: Conference CA Section: 1 (Pharmacology) Sulpiride [15676-16-1] (100 mg, i.v.) increased plasma noradrenaline (NA) [51-41-2] in healthy volunteers which was more pronounced with 50 mg i.v. under ergometric charges. Maprotiline [10262-69-8] produced a significant increase, but desimipramine [50-47-5] a significant decrease of NA plasma levels with 25 mg i.v. under ergometric work although both substances are powerful NA reuptake inhibitors. After mianserine [24219-97-4] (1 mg, i.v.) there was a significant increase of NA plasma concn. under resting conditions, but not with ergometric charges. The results are discussed in relation to the mechanism of action of the different compds. and to their antidepressant properties.
