DOCUMENT TYPE: Journal CA Section: 17 (Food and Feed Chemistry) Section cross-reference(s): 4 The Na [128-44-9], K [10332-51-1], Ca [6485-34-3], and Mg [88092-79-9] salts of saccharin, and MgCl2, were clastogenic to Chinese hamster lung fibroblasts in vitro, but only at elevated doses (8-16 mg/mL). Saccharin acid [81-07-2] was inactive to the limits of its soly. (4 mg/mL). When the data are expressed in terms of ionic concn., each salt showed a similar clastogenic potency. Thus, ionic effects induced by these salts in the assay medium may be the crit. determinant of the clastogenic effects seen, rather than the saccharin moiety presenting a genotoxic insult to the chromosomes of the cells. The metal-chelating agents EDTA [60-00-4] and EGTA [67-42-5] were nonclastogenic, but disodium EDTA [139-33-3] showed weak activity prior to toxicity at 0.5 mg/mL. The absence of a clastogenic response to the results of saccharin at doses of <4 mg/mL is discussed within the context of the threshold-dependent tumor-promoting activity of high dose levels of Na saccharin to the bladder of male rats. The doubtful value of conducting in vitro clastogenicity studies at doses of >10-2M is discussed.
