DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) In isolated hearts of rabbits, perfusion with amitriptyline-HCl [549-18-8], maprotiline-HCl [10347-81-6] (4.8.mu.M), or mianserin-HCl [21535-47-7] (28.8.mu.M) reduced ventricular force but did not change heart rate; only amitriptyline reduced atrial force and lengthened the interval between peak atrial and peak ventricular tensions (A-V contraction interval). At higher concns. (amitryptyline 8, maprotiline 8, and mianserin 60.mu.M), all the agents produced dysrhythmias interfering with atrioventricular synchronization. In the presence of mianserin (4.8.mu.M), perfusion with (-)-noradrenaline bitartrate [51-40-1] (0.0059-5.9.mu.M) shortened the A-V contraction interval but did not produce dysrhythmias. In the presence of amitriptyline, maprotiline (4.8.mu.M), or mianserin (28.8.mu.M), noradrenaline perfusion results in the lengthening of the A-V contraction interval and dysrhythmias. 3H-labeled noradrenaline uptake in perfused rabbit hearts and mouse isolated atria or vasa deferentia was inhibited by the antidepressants to a similar extent. Predisposition to noradrenaline-induced dysrhythmias produced by the antidepressants was not related to blockade of noradrenaline uptake.
