DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) (+)-Amphetamine sulfate (I) [51-63-8] (5 ′ 10-5-10-2M), perfused for 5 min into the mesencephalic reticular formation (MRF) of cat encephale isole prepns., mimicked the electrocortical desynchronization induced by (-)-noradrenaline-HCl (II) [329-56-6] or (-)-a-methylnoradrenaline-HCl (III) [10390-18-8] at the same site. Perfusion of 6-hydroxydopamine-HCl (IV) [28094-15-7] also induced desynchronization, but, over the 1-h perfusion, the slow wave activity returned to the elec. record. Pretreatment with IV abolished the effect of I but not the effects of II and III; pretreatment with II or III restored the blocked effect of I. Fluorescence studies with III indicated that IV depleted noradrenergic nerve terminals near the cannulae tips, but these terminals were still capable of taking up exogenous III. I may have a presynaptic action on noradrenergic nerve terminals within the MRF.
