DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) The liver post-mitochondrial supernatants (S-9 mix) from 5 species including man (1 accident victim) were tested for activating activity with dimethylnitrosamine (I) [62-75-9], methylethylnitrosamine (II) [10595-95-6], dipropylnitrosamine (III) [621-64-7], nitrosopiperidine (IV) [100-75-4], and nitrosopyrrolidine (V) [930-55-2] in the Ames test; with mouse and hamster S-9 mix activating I, II, and III, but human, rat, and pig S-9 mix not. IV was activated by all species and V was activated by all but the rat; human S-9 mix was exceptionally activating for V. Enzyme levels in hepatic microsomes of all 5 species were detd. and had no relation to S-9 mix metabolic activation ability.

DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) Dimethylnitrosamine [62-75-9], dipropylnitrosamine [621-64-7], methylethylnitrosamine [10595-95-6], nitrosopiperidine [100-75-4], and nitrosopyrrolidine (I) [930-55-2] were assayed for mutagenicity in the Ames test in the presence of hepatic postmitochondrial prepns. isolated from the mouse, rat, hamster, pig, and man. Prior to each mutagenicity assay all activation systems were fully characterized with respect to monoamine oxidase [9001-66-5], mixed-function amine oxidase [9076-86-2], and mixed-function oxidase [9040-60-2] activities. The hamster was the most efficient activator for all nitrosamines followed by the mouse. The latter species, however, activated I only weakly which was the only carcinogen readily activated by the human prepn. None of the aliph. nitrosamines was activated by the rat or pig. No correlation was obsd. between efficiency of activation and any of the enzyme activities studied.
