DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Male rats weighing 100 g were injected daily for 10 days with vehicle (control) or with 0.4, 2.0, 4.0, 10.0, or 20.0 mg/kg/day of ethane-1-hydroxy-1,1-diphosphonate (I) [2809-21-4] or dichloromethylene diphosphonate (II) [10596-23-3]. All doses of II and doses of .gtoreq.2.0 mg I/kg/day increased the percentage of hard tissues, with II being more effective than similar doses of I in decreasing bone resorption. Osteoclast population parameters were increased with all doses of both I and II, with II having a greater effect than similar doses of I. Decreases in the proliferation of the osteoprogenitor pool paralleled the decreases in osteoblasts and bone formation parameters. These decreases in osteoprogenitor pool proliferation did not account for the increases with diphosphonates in osteoclast population parameters.

DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) In (dichloromethylene)diphosphonic acid (I) [10596-23-3]-osteopetrotic mice, one subpopulation of thioglycollate-induced peritoneal exudate macrophages (Mo) is missing. This subpopulation is the one whose differentiation is dependent on T-lymphocytes, as it is also missing in the athymic nu/nu mice. I-induced osteopetrosis, being partially attributable to deficient osteoclastic bone resorption, suggests that this missing Mo subpopulation might represent the precursors of osteoclasts. Thus, the interplays between T-cells and Mo, so well known in immunity and inflammation, may also be relevant to osteoclastic differentiation and therefore, to bone remodeling.
