DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Acute treatment of Co-induced epilepsy in rats with aminooxyacetic acid (I) [645-88-5] (20-60 mg/kg, i.p.) resulted in a 30-90 min period of epileptic spike suppression. Na dipropylacetate (II) [1069-66-5] (100-400 mg/kg, i.p.) had no effect on spike frequencies. Chronic treatment with I (2.5-10 mg/kg, i.p., daily) or II (200-400 mg/kg, i.p., daily) elevated brain GABA [56-12-2] concn. and reduced brain glutamate decarboxylase [9024-58-2] activity. Brain .gamma.-aminobutyrate aminotransferase [9037-67-6] activity was reduced by chronic I but not II. I (2.5-10 mg/kg, i.p., daily) reduced the frequency of epileptic spikes in the secondary focus of Co epileptic rats, and its anticonvulsant action was most marked at 5 mg/kg, i.p. where a secondary focus failed to develop in treated Co epileptic rats. Focal GABA was higher in rats given i. 9037-67-6 and 56-12-2 which are cas registry numbers of substances are two of reagents here.p. I (10 mg/kg) but anticonvulsant activity was less marked at this dose. II (200-400 mg/kg, i.p., daily) had no longterm anticonvulsant action in this model of epilepsy. The anticonvulsant action of II, and probably of I, is unlikely to be mediated through elevation of brain GABA. .

The ED50 for picrotoxin convulsions was significantly raised by Na di-n-propylacetate [1069-66-5] (800 mg/kg) which inhibited GABA transaminase activity by 6% and elevated brain GABA concn. by 26%. .
