DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) The change of n-hexane [110-54-3] metabolites in the urine of rats exposed to various concns. of n-hexane and to its mixt. with toluene [108-88-3] or MEK [78-93-3] was studied. In the 1st expt., 5 sep. groups of 5 rats each were exposed to 100, 500, 1000, or 3000 ppm n-hexane or fresh air, resp., in an exposure chamber for 8 h/day. Urinary samples were gathered during exposure and 16, 24, and 40 h after exposure. Half of each sample was analyzed by gas chromatog. after hydrolysis with acid and enzymes, and the other half was analyzed without hydrolysis. 2,5-Dimethylfuran [625-86-5], MBK [55-48-1], 2-hexanol [626-93-7], 2,5-hexanedione [110-13-4], and g-valerolactone [108-29-2] could be identified as n-hexane metabolites in the urine. The main metabolites were 2-hexanol and 2,5-hexanedione. 2-Hexanol was mostly excreted during exposure, while most of the 2,5-hexanedione was excreted after the end of exposure. The amt. of metabolites in the urine correlatively increased with the concn. of n-hexane from 100 to 1000 ppm, but the amt. of metabolites scarcely increased when the concn. of n-hexane increased from 1000 to 3000 ppm. The max. concn. of the excreted metabolites in urine was delayed when the concn. 108-88-3 and 78-93-3 are also in the experiment. of n-hexane increased. Thus, the capacity of hydroxylation in rat liver microsomes was satd. when exposed to high concns. of n-hexane. Free (not conjugated) metabolites were also excreted in the urine in all concns. during exposure and 16 h after exposure. In the 2nd expt., 4 sep. groups of 5 rats each were exposed to 1000 ppm n-hexane, 1000 ppm n-hexane plus 1000 ppm toluene, 1000 ppm n-hexane plus 1000 ppm MEK, or fresh air. The analyzing procedure was the same as that in the 1st expt. Distributions of n-hexane metabolites in the mixed exposure group were almost similar to that of the n-hexane-alone group. The amt. of metabolites decreased to ~1/6 of that in the n-hexane group by co-exposure with toluene and to ~1/4 by co-exposure with MEK. Toluene decreases the neurotoxicity of n-hexane by the inhibition of n-hexane metab. However, the reason the neurotoxicity of n-hexane is increased by co-exposure with MEK cannot be clearly explained. .
