DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Dietary digitonin (I) [11024-24-1] was well tolerated by rats and cynomolgus monkeys, and prevented the expected rise in plasma cholesterol [57-88-5] in monkeys fed a diet contg. butter and cholesterol. Both I and mildly hydrolyzed I pptd. cholesterol dispersed in micellar suspensions and decreased intestinal absorption of cholesterol in rats, but only I bound cholesterol in vitro. The hypocholesterolemic effect of I may be secondary to decreased intestinal absorption of cholesterol assocd. with a disturbance in the micellar suspension of cholesterol in intestinal contents.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) In concns. greatly exceeding those of the hemolysin, glycyrrhizin (I) [1405-86-3] inhibited the hemolytic activity of saponins, e.g. escin [6805-41-0], on bovine erythrocytes in vitro. There was a direct relation between the concn. of each saponin used and the concn. of I required for 50% protection. I may prevent access of the hemolysin to its erythrocyte receptor. In rats, 70 mg I/kg was toxic. When injected with digitonin [11024-24-1] (LD50 23 mg/kg, i.v.) 30 mg I/kg did not affect mortality, but 50 mg I/kg reduced mortality by 40%. Neither I dose reduced the hemolysis, possibly due to the low ratio of inhibitor to hemolysin in vivo.
