DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) The binding of 125I-labeled a-bungarotoxin (I) [11032-79-4] to rat muscle sarcolemma membranes was increased ￡2.4-fold by adding phospholipase C [9001-86-9] (from Staphylococcus aureus) to the incubation medium; 4b-phorbol 12b,13a-dibutyrate [37558-16-0] also increased I binding, thus mimicking the effect of phospholipase c. A diglyceride product, formed during hydrolysis by the enzyme, could be a 2nd messenger for the receptor binding sites.

P15 toxin, a probe for ganglionic nicotinic receptor, prevents the binding of a-Bgtx to rat ganglia, and a-Bgtx prevents the ganglioplegic action of hexamethonium. Hexamethonium does not block the binding of a-Bgtx and P15 to rat ganglia. Thus, a-Bgtx and P15 bind to ganglionic nicotinic receptor at a common site which is different from that of cholinergic agonists and antagonists.
