DOCUMENT TYPE: Journal CA Section: 43 (Cellulose, Lignin, Paper, and Other Wood Products) In the soda pulping of beech and spruce holocellulose (I) [8064-26-4], the addn. of quinone (II) derivs., i.e., anthraquinone (III) [84-65-1], 1,4,4a,9a-tetrahydroanthraquinone (IV) [56136-14-2] and their derivs., stabilized the carbohydrates, and the amt. of carbohydrate stabilized in spruce I was nearly the same as that in beech. The stabilization of carbohydrates by II derivs. was attributed to the prevention of degrdn. of cellulose (V) [9004-34-6] and glucomannan (VI) [11078-31-2] in spruce and V and xylan (VII) [9014-63-5] in beech I; the V was effectively stabilized by III and IV while VI and VII were stabilized more effectively by III and IV derivs. than by III and IV. The redn. in d.p. of V was suppressed by addn. of II derivs. .

DOCUMENT TYPE: Journal CA Section: 9 (Biochemical Methods) Section cross-reference(s): 33 Interaction between aniline blue and curdlan, a (1?3)-b-D-glucan, was studied by using absorption and fluorescence spectroscopy. The evidence suggests that a minor, weakly fluorescent component of com. dyes forms a strongly fluorescent complex with curdlan, with an excitation max. of 395 nm and an emission max. of 495 nm. This component was partially purified by TLC on silica gel. Of many polysaccharides surveyed, a no. showed weak interactions with the major component of aniline blue but only (1?3)-b-D-glucans such as pachyman, curdlan and laminaran induced fluorescence in the minor component. 11078-31-2 and 9005-25-8 are also in the experiment. Fluorescence was less with laminaran than with curdlan and decreased with increasing alkali concn. suggesting conformation control of the dye-binding mechanism. As little as 5 mg/mL of curdlan induced easily detectable fluorescence increases in aniline blue, and this was used to demonstrate the presence of (1?3)-b-D-glucan in a fungal cell wall prepn. (1?3)-b-D-Glucan in cereal grain sections was located as bright yellow-green fluorescent particles. In barley these stained particles, located in assocn. with the subaleurone endosperm cell wall, showed a fluorescence excitation max. at 395 nm and emission max. of 495 nm. .
