DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) Aroclor 5432 [63496-31-1], Aroclor 5442 [12642-23-8], and Aroclor 5460 [11126-42-4] administered to mice by gastric intubation at 250, 500, and 1000 mg/kg lowered in vitro D-glucose [50-99-7] intestinal absorption. The absorption was greater, although not significantly, after treatment with the polychlorinated terphenyls (PCT's) with higher Cl content. Using D-glucose as the substrate neither serosal fluid transfer nor gut fluid uptake were altered by 50-1000 mg PCT/kg. D-Galactose, L-arginine, and L-histidine absorption and mucosal fluid transfer were unaltered by the PCT's. The addn. of D-mannose [3458-28-4], an exogenous energy supply, increased both D-glucose absorption and total fluid transfer in PCT-treated animals (however, both parameters were virtually unchanged compared to untreated controls with an added energy supply).

DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) Exposure of male rats to the polychlorinated terphenyl (PCT) mixts. Aroclor 5460 [11126-42-4] and Aroclor 5432 [63496-31-1] contg. 60% and 32% (wt./wt.) of Cl, resp., showed that the PCT mixt. with a low degree of chlorination, Aroclor 5432, was a potent inducer of liver microsomal cytochrome P 450 [9035-51-2], aryl hydrocarbon hydroxylase (AHH) [9037-52-9] and 7-ethoxyresorufin O-deethylase [59793-97-4] whereas Aroclor 5460 and the unchlorinated isomers o- [84-15-1], m- [92-06-8], and p-terphenyl (I) [92-94-4] were weak inducers.Several substances with their cas registry numbers 92-94-4 and 84-15-1 may be metioned in this study. Ultrastructurally, proliferation of smooth but not rough endoplasmic reticulum, frequent occurrence of lysosomes contg. partially degraded lipid material as well as an increased no. and size of cytoplasmic lipid droplets were obsd. These changes were most pronounced in Aroclor 5432-treated rats. Competition expts. with [3H]TCDD for binding to the cytosolic receptor protein indicated the presence of Aroclor 5432, but not in Aroclor 5460, of components with receptor affinity. These components represent only a minor fraction of the mixt. as judged by the 1900-fold excess necessary to displace 50% of the specific [3H]TCDD binding. Thus, the PCT mixt. Aroclor 5432 is a mixed type inducer of hepatic cytochrome P 450 in the rat. The presence of Aroclor 5432 of compds. capable of inducing AHH in vivo and of binding to the TCDD-receptor might be highly relevant with regard to the potential toxicity in view of the apparent correlation between affinity for the TCDD-receptor, induction of AHH activity and toxic properties for chlorinated arom. hydrocarbons. .
