DOCUMENT TYPE: Patent CA Section: 16 (Fermentation and Bioindustrial Chemistry) Xanthan gum (I) [11138-66-2] is produced by culturing Xanthomonas campestris NRRL B-1459 in a medium contg. fumaric acid [110-17-8], malic acid [6915-15-7], uracil [66-22-8], riboflavin [83-88-5], or their salts, esters, or derivs. Thus, X. campestris NRRL B-1459 was cultured with shaking at 28° for 95 h in 100 mL medium contg. glucose 20, KH2PO4 5, MgSO4.7H20 0.2, urea 2.7 g/L, and 0.5% Na fumarate. The culture broth was dild. 3 times with H2O and centrifuged. The supernatant was mixed with 3 g KCl and 400 mL MeOH to ppt. I, which was washed with 100 mL MeOH and dried under vacuum. The yield of I with a viscosity of 4420 cP (1% soln.) was 12.71 g/L, whereas 11.39 g/L of I with a viscosity of 1310 cP (1% soln.) was obtained cultured without addn. of Na fumarate.

Thus, in a lab. expt., X. cannabis ICPP XC12 was cultured on a medium contg. 2% sucrose, 0.2% yeast ext., 0.2% (NH4)2SO4, 1.15% Na2HPO4, 0.03% KH2PO4, 0.1% NaCl, 0.6% soybean flour, and 0.06% rapeseed oil, at pH 7.9 and 30°. After 64 h, the xanthan gum yield was 14.1 g/L.
