DOCUMENT TYPE: Journal CA Section: 64 (Pharmaceutical Analysis) A simple, sensitive and accurate spectrophotometric method for the detn. of metformin hydrochloride (MfCl) [1115-70-4] is described. The method is based on the formation of charge transfer complex (C.T.) with iodine in acetonitrile medium. Beer's law is obeyed within the concn. range 1.656-72.86 mg/mL of MfCl whereas the optimum concn. range as evaluated by Ringbom's method is 6.62-66.2 mg/mL. The formation const. of the formed complex is evaluated and was found to be 3.8 ′ 104 L mol-1. The molar absorptivity, Sandell sensitivity, relative std. deviation and recovery percent are evaluated. The results are treated statistically. Applying the F- and t-tests and found highly accurate, precise and reproducible. The method is used to det. MfCl in pure form and pharmaceutical prepns. There is no interference from the excipients.

For example, a compn. was prepd. contg. TEFOS 1500 10, stearic acid 1, liquid petrolatum 8, galactomannan gel 30, metformin-HCl [1115-70-4] 5.33, and deionized water 45.67 g. Twice daily administration of the above compn. for 30 days caused a marked and rapid treatment of a previously incurable cellulitis in a female patient.
