Isolation and characterization of mildiomycin, a new nucleoside antibiotic. Harada, Setsuo; Kishi, Toyokazu (Cent. Res. Div., Takeda Chem. Ind., Ltd., Osaka, Japan). J. DOCUMENT TYPE: Journal CA Section: 16 (Fermentations) A new antibiotic mildiomycin [67527-71-3], strongly active against powdery mildew, was isolated from the culture filtrate of Streptoverticillium rimofaciens B-98891. It is a water-sol. basic antibiotic and was purified by ion-exchange and adsorption chromatog. The mol. formula of the purified compd. was detd. to be C19H30N8O9(H2O) from phys. and chem. data. The UV and NMR spectra suggested that this antibiotic is a nucleoside. On acidic hydrolysis it gave 5-hydroxymethyl cytosine [1123-95-1] which has not previously been found in nucleoside antibiotics.

DOCUMENT TYPE: Journal CA Section: 16 (Fermentation and Bioindustrial Chemistry) Section cross-reference(s): 5 Mildiomycin (I) [67527-71-3], produced by Streptoverticillium rimofaciens, is an antibiotic with a 5-hydroxymethylcytosine (HMC) [1123-95-1] moiety. HMC, when added into the fermn. medium, promoted the prodn. of I. However, cytosine [71-30-7] suppressed I prodn.; a I analog (II [78162-87-5]) having a cytosine instead of the HMG moiety of I was produced, together with a significant amt. of HMC. In the same way, the 5-methyl-, 5-iodo-, and 5-fluorocytosine analogs of I were formed and I prodn. was suppressed. These I analogs showed strong antifungal activities against powdery mildew.
