DOCUMENT TYPE: Journal CA Section: 17 (Food and Feed Chemistry) A simple, rapid, and sensitive anal. method was developed for the detn. of 2 fluoroquinolones, danofloxacin and marbofloxacin, in bovine milk samples. Sepn. and quantification were performed by micellar liq. chromatog. with fluorescence detection (MLC-FD), using sodium dodecyl sulfate (SDS) as a surfactant. The influence of the principal factors, namely, the micelle concn., the amt. of org. modifier, tail-reducing agents, the pH, and the temp. were studied. The suitable condition was found to be 75 mM SDS-10 mM phosphate buffer-18 mM tetrabutylammonium bromide/3% (vol./vol.) 1-propanol at pH 3.0 for the sepn. of marbofloxacin, danofloxacin, and tosufloxacin (internal std.) in about 20 min. The linear concn. range of application was 1.8-30.0 ng × mL-1 for danofloxacin and 16-120 ng × mL-1 for marbofloxacin, and the relative std. deviation ranged between 4.9 and 2.7%. The limit of detection found for danofloxacin was 0.5 ng × mL-1 and 5 ng × mL-1 for marbofloxacin.There are some commonly used reagents with their cas registry numbers 115550-35-1 and 112398-08-0 in this article. These values were lower than the max. residue limits (MRLs) established by the European Union for these compds. in bovine milk. It was applied to check the eventual existence of these compds. above these limits on com. milk samples. The validation method was completed with spiked milk samples. Recovery levels obtained were 90.3-108.2%. .

Samples were extd. with a mix of acetonitrile and 0.1 M citrate, 150 mM MgCl2, pH 5.0. After centrifugation and evapn., the exts. could be analyzed by liq. chromatog. with fluorescence detection. Good recoveries (63-95%) were obtained from samples fortified with a mix of 5 fluoroquinolones and 3 tetracyclines, with satisfactory relative std. deviations. Limits of detection were 0.5 ng/g (danofloxacin), 1 ng/g (oxytetracycline, ciprofloxacin, enrofloxacin), 1.5 ng/g (tetracycline), 2 ng/g (difloxacin) and 5 ng/g (sarafloxacin, chlortetracycline). Enrofloxacin and its metabolite ciprofloxacin, as well as oxytetracycline were detd. in enrofloxacin and oxytetracycline incurred chicken muscle using this method. .
