DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) 8-Arginine-vasotocin (AVT) [113-80-4], oxytocin [50-56-6], and 8-arginine-vasopressin (AVP) [113-79-1] were injected intracerebroventricularly (i.c.v.) and(or) into the hypothalamus of awake, freely moving rats to investigate the potential role of these neuropeptides in central cardiovascular control. AVT (0.015-10 nmol) injected i.c.v. caused dose-dependent pressor responses; low doses also elicited tachycardia, whereas the higher doses (1 and 10 nmol) produced bradycardia. In contrast, oxytocin injections into the lateral ventricle (0.015-100 nmol) had no cardiovascular effects. Plasma vasopressin was not affected by administration of oxytocin or AVT except at the highest dose of AVT. AVT (0.1 or 1 nmol) and AVP (0.015 or 0. 113-79-1 and 50-56-6 which are cas registry numbers of substances are two of reagents here.1 nmol) injected into the hypothalamic nucleus preopticus medialis (POM) increased blood pressure and heart rate; these changes were much greater than with comparable doses of AVT injected i.c.v. Plasma levels of norepinephrine [51-43-4] and epinephrine [51-41-2], but not vasopressin, were elevated during the pressor period induced by 1 nmol of AVT injected into the POM. The pressor and cardiac accelerating effects of AVT (1 nmol) injected into the POM were diminished in bilaterally adrenal demedullated, bretylium-treated rats. The data suggest a role for AVT, but not oxytocin, in central cardiovascular control through activation on the sympathoadrenomedullary axis. The anteroventral hypothalamus might play a central role in these autonomic effects of vasotocin. Evidently, AVP and AVT share some central autonomic effects due to structural similarities. .

DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) The cAMP [60-92-4] level in isolated frog skin epithelium was stimulated by a range of concns. of 8-arginine vasotocin (AVT) [113-80-4] and compared to active Na transport measured as short-circuit current (SCC). The response of SCC and osmotic H2O flow (OWF) to AVT was investigated in sep. expts. The SCC was ~3 times more sensitive to AVT than was OWF. Measurable increments of cAMP above the basal level were found only with AVT concns. eliciting greater than the half-maximal response of SCC. Two models are offered to explain the findings: (A) total SCC depends on epithelial cAMP with a sigmoidal relation and (B) epithelial cAMP exists in 2 sep. pools of which only 1 is accessible to AVT, and SCC stimulation depends on cAMP in the AVT-controlled pool in a simple saturable fashion. Correlation between cAMP and SCC after stimulation with theophylline resembled that after AVT. Papaverine [58-74-2] (10 mM) induced only small changes in SCC inspite of a substantial increase in cAMP level. 58-74-2 and 60-92-4 which are cas registry numbers of substances are two of reagents here. Higher concns. of papaverine inhibited both basal and AVT-stimulated SCC, but the cAMP level was further increased. This effect of papaverine may be due to a simultaneous block of another rate-detg. process. Papaverine had no effect on basal or AVT-stimulated OWF despite a marked stimulation of the cAMP level. Thus, the role of cAMP as mediator of the hydroosmotic action of AVT must be questioned. .
