Thus, 200 mL of a preculture of S. faecalis ATCC 8043 was incubated in 20 L of a pH 6.8 medium contg. casamino acids, bactotryptone, L-cysteine, L-aspartic acid, DL-alanine, DL-tryptophan, glucose, Na acetate, Na succinate, KH2PO4, K2HPO4, FeSO4.7H2O, MnSO4.H2O, MgSO4.7H2O, NaCl, thiamin, riboflavin, pyridoxine, pyridoxal, niacin, Ca pantothenate, folic acid, p-aminobenzoic acid, biotin, adenine sulfate, guanine-HCl, uracil, and Tween 80 at 37° for 16 h and centrifuged. The cell mass was lysed by sonication, centrifuged, and a crude I prepn. was obtained by pptn. with (NH4)2SO4. The crude I was purified ~19,000-fold at 115% yield by chromatog. on DEAE-Sephadex A-50, Sephadox G-150, AH-Sepharose 4B, Sepharose 6B, and hydroxyapatite. The pH and temp. optima of purified I were 6.0 and 35°, resp. I reacted with N-a-carbobenzoxyglycine [1138-80-3] to produce glycine [56-40-6] and with N-a-carbobenzoxyalanine [1142-20-7] to produce benzyl alc. [100-51-6] and alanine [56-41-7].

Treatment of BzOH with I gave the benzoyloxazolone II, which reacted with HOCH2CH2SH in the presence of Et3N to give 63% HOCH2CH2SBz. Treatment of MeNO2 with II in THF contg. diazabicycloundecene gave 59% O2NCH2COPh. .
