DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) Three mature Large White boars were anesthetized and received 7(n)-3H-labeled pregnenolone [145-13-1] by continuous infusion into right and left spermatic arteries for up to 180 min. Spermatic venous blood flow was measured by sep. timed collections of completely diverted outflow from each testis, and blood not sampled was returned to the peripheral circulation. The total radioactivity in plasma from each testis increased markedly during the 1st 60 min of infusion to reach a plateau at 80-180 min. Radiolabeling of 5a-androst-16-en-3-one [18339-16-7], 5a-androst-16-en-3b-ol [7148-51-8], and 5a-androst-16-en-3a-ol [1153-51-1] showed similar patterns with ratios of mean radioactivity of 5:3:1 between 80 and 180 min. In comparison, the amts. of tritiated 4,16-androstadien-3-one [4075-07-4] formed were very small. The radiolabeling of testosterone [58-22-0] and 4-androstenedione [63-05-8] occurred more rapidly than that of the 16-androstenes and reached max. by 30 min. However, the amts. were only 20% (testosterone) and 10% (4-androstenedione) those of the combined quantities of tritiated 16-androstenes. Addn. of human chorionic gonadotropin (hCG) [9002-61-3] to the infusate to 1 testis in each animal (so that 5000 IU hCG were delivered in 15-20 min) produced no change in the outputs of radiolabeled steroids although RIA of spermatic venous plasma in samples from one expt. showed a transient increase in the concn. of 4-androstene-3,17-dione [63-05-8] during the hCG infusion. The lack of response to hCG could be produced by satn. and down-regulation of binding sites by the very high local concns. of hCG.

DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) The metab. of plasma 3H-labeled androstenone (I) [18339-16-7] administered i.v. was studied in young boars weighing ~100 kg. The total radioactivity per L of plasma increased up to 14 min after the injection, and then declined rather slowly; plasma radioactivity was still measurable 7 days after injection. The metabolic clearance rate of I was ~80,000 L/day. This quick disappearance of plasma I was probably due mainly to storage in fatty tissue and, to a lesser extent, to catabolism to 5a-androst-16-en-3a-ol [1153-51-1], 5a-androst-16-en-3b-ol [7148-51-8], and esp. to at least 3 unknown more polar compds. Radioactivity was mainly eliminated in the urine in the form of the same unknown polar compds.
