DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) The possible toxicity and carcinogenicity of a 5-10% soln. of p-amino-o-nitrophenol [119-34-6], a 5-10% soln. of p-phenylenediamine [106-50-3] and a 1-2% soln. of Na thioglycollate [367-51-1] on mouse and rabbit skin were studied by twice-weekly topical application of these compds. (0.02 mL/animal) in acetone soln. There was no decrease in the lifespan of either species. The mice suffered no marked wt. loss, and no abnormalities were found in the blood or urine of rabbits. Untreated as well as treated mice presented a large no. of tumors, including lymphomas, lung adenomas, liver hemangiomas and ovarian neoplasms; dermal fibromas, but no epidermal tumors, were found in all the treated mice. No increase in tumor incidence was found, however, in a comparison of the treated mice with untreated controls, and no neoplasms were obsd. in the rabbits. In pos. controls of both species, large nos. of skin tumors were induced by small doses of 7,12-dimethylbenz[a]anthracene, confirming the sensitivity of these models for skin carcinogenesis studies.

DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) Section cross-reference(s): 62 A dominant lethal mutagenicity study was performed in rats with the following chems. that may be used to dye hair: 2-nitro-p-phenylenediamine (I) [5307-14-2], 4-nitro-o-phenylenediamine [99-56-9], m-phenylenediamine [108-45-2], o-phenylenediamine [95-54-5], p-phenylenediamine [106-50-3], p-toluenediamine [95-70-5], 2,4-diaminoanisole [615-05-4], 2,5-diaminoanisole [5307-02-8], 2-amino-4-nitrophenol [99-57-0], 2-amino-5-nitrophenol [121-88-0], and 4-amino-2-nitrophenol [119-34-6]. The compds. were administered i.p. 3 times weekly for 8 wk to groups of 20 sexually mature Charles River CD male rats at a dose of 20 mg/kg. This amt. of dye is an enormous exaggeration of the human exposure from brief monthly topical application of a hair color product contg. 2 g (40 mg/kg) or less total dye. There was no evidence of an increase in postimplantation fetal loss which could indicate a dominant lethal effect. Included are data on the acute toxicity of the dyes by various routes of administration in different vehicles.
