DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Acetylacetonate-1,5-cyclooctadienerhodium(I) (I) [12245-39-5] and cis-dichlorotetrakisdimethylsulfoxide ruthenium(II) (II) [64376-67-6] were examd. for antitumor activity. I had an activity comparable with that of cis-diamminedichloroplatinum(II) (III) against Ehrlich ascites carcinoma in mice, yielding 100% cures at 25 or 50 mg/kg/day i.p. The comparative activity was slightly lower. Only II and III had any activity against L1210 leukemia. The histol. damage caused by the compds. tested was moderate and was virtually absent for I on intestinal mucosa and spleen. I caused a selective depression of labeled uridine incorporation into tumor cells. II caused a similar inhibition of incorporation of thymidine, uridine, and leucine at the highest dosage tested, but not at lower dosages that were still therapeutically useful. Thus I and II have a therapeutic efficacy better than or equal to that displayed by III, and they may have a different mode of action.

The electronic effects on the outcome of the enantioselective catalysis have been analyzed. ..
