DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) The pharmacokinetics and pharmacodynamic effects of i.v. injected biosynthetic human insulin [11061-68-0] of recombinant DNA origin and purified pig insulin [12584-58-6] were compared in healthy volunteers. Both had identical immunol. properties. Some of their pharmacokinetic features, such as total plasma clearance and bioavailability, were similar, but they appeared to differ in biol. half-life, elimination rate const., and apparent vol. of distribution. There was no appreciable difference between their hypoglycemic activity and their inhibitory effect on endogenous insulin secretion. The glucagon [9007-92-5]-secretory response seemed to be less pronounced after biosynthetic human insulin than after porcine insulin. .

DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) The relative potencies of porcine insulin [12584-58-6] and semisynthetic human insulin [11061-68-0] were examd. in humans using the euglycemic glucose [50-99-7] clamp technique. Three 90-min, consecutive infusions at 2.4, 4.8, and 7.2 units/h were administered to 7 normal subjects. The rate of glucose infusion required to maintain euglycemia during the last 40 min of each insulin infusion for porcine (P) and human (H) insulin was not different. However, when the glucose infusion rate was expressed as a function of the mean serum insulin level over the same period of time the values for P and H at 4.8 and 7.2 units/h were not significantly different but H was greater than P. Mean serum insulin levels did not differ for human or porcine insulin infusion. This study confirms the similarity in potency between human and porcine insulin in the human. However, delivery at 2.4 units/h, which would predominantly affect hepatic glucose output, suggests that semisynthetic human insulin is more potent than porcine insulin with respect to the liver. .
