CLASS: IC: A61K009-14; A61K047-00. APPLICATION: JP 82-114083 2 Jul 1982. DOCUMENT TYPE: Patent CA Section: 63 (Pharmaceuticals) The soly. of barely sol. drugs is increased when the drugs are pulverized with chitin [1398-61-4] or chitosan [9012-76-4] and their particle size decreased in a ball milling app. Thus, a griseofulvin [126-07-8] and 200-mesh chitosan mixt. (1:9) was pulverized 24 h, mixed with lactose, and made into tablets. .

- Ultrastructural changes of intrahepatic bile ductiles in griseofulvin fed mice. Shapiro, Stanley H.; Wessely, Zelma (Queens Hosp. Cent., Long Island Jewish-Hillside Med. Cent., Jamaica, NY 11432, USA). Ann. Clin. 64-17-5 and 126-07-8 are cas registry numbers. These chemicals are also mentioned in this article. Lab. Sci., 14(1), 69-77 (English) 1984. CODEN: ACLSCP. ISSN: 0091-7370. DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Section cross-reference(s): 14 Intraphepatic bile ductules in male mice, fed a diet contg. 2.5 percent griseofulvin (GF) [126-07-8], showed marked hypertrophy and hyperplasia within 2 wk. Thereafter, this reaction gradually increased in prominence until 9 wk. Cytoplasmic acid phosphatase and succinic dehydrogenase deposits were more prominent and abundant after 12 to 14 wk. Ultrastructural changes in ductular epithelial cells included enlargement of nuclei and nucleoli and increased abundance of cytoplasmic rough surfaced endoplasmic reticulum, ribosomes and mitochondria until 9 to 12 wk. At this interval, duct lumina appeared widely dilated with small clumps of electron dense material at the periphery adjoining apical cell membranes with severely flattened or absent microvilli. Electron dense needle-like crystals resembling protoporphyrin were clustered centrally in the lumina. After 12 wk occasional mitochondria were markedly swollen and contained vacuolated areas. Numerous multivesicular bodies were noted. Mallory bodies were seen in a few duct cells. The spectrum of duct cell changes just noted gives further evidence of extensive liver injury after GF feeding. These changes, in addn. to diverse hepatocyte alterations previously reported, constitute morphol. features of GF-induced murine porphyria, also common to human porphyria and human alc. liver disease. .
