DOCUMENT TYPE: Journal CA Section: 2 (Hormone Pharmacology) Injection of rats with 5.alpha.-pregnane-3.beta.-ol-20-one (I) [516-55-2], 5.beta.-pregnane-3.alpha.-ol-20-one (II) [128-20-1], 5.alpha.-pregnane-3.beta.,20.beta.-diol (III) [516-53-0], and 5.beta.-pregnane-3.alpha.,20.alpha.-diol (IV) [80-92-2] (20 mg/kg, s.c., daily for 7 days) reduced the activities of aminopyrine N-demethylase [9037-69-8], coumarin 3-hydroxylase [9045-41-4], and glucose 6-phosphatase [9001-39-2] in hepatic microsomes. 16.alpha.-Hydroxyprogesterone (V) [438-07-3] and pregnenolone-16.alpha.-carbonitrile (VI) [1434-54-4] (20 mg/kg, s.c., daily for 7 days) increased drug metab. and glucose 6-phosphatase activity. I, II, III, and IV decreased the synthesis of microsomal phospholipids, and reduced the activity of methyltransferase [9033-25-4] and the total microsomal phosphatidylcholine (PC):phosphatidylethanolamine (PE) ratio. V and VI increased microsomal phospholipid synthesis, methyltransferase activity, and the PC:PE ratio. I, II, III, and IV decreased microsomal progesterone [57-83-0] hydroxylation in the 16.alpha.- and 6.beta.-position whereas V and VI increased it. Of the steroids tested in concns. of 1.3 ′ 10-6M and with incubation times from 0-24 h, progesterone, estradiol, and 11-ketotestosterone were ineffective, whereas 17a-hydroxyprogesterone [68-96-2], 20b-dihydroprogesterone [145-15-3], 17a-hydroxy-20b-dihydroprogesterone [1662-06-2], epipregnanolone [128-20-1], testosterone [58-22-0], and hydrocortisone (I) [50-23-7] were effective in stimulating GVBD after 3 h, estrone [53-16-7] after 1 h, and 11-deoxycorticosterone acetate (II) [56-47-3] after 20 min. Estrone, 17a-hydroxy-20b-dihydroprogesterone, and II were also effective in concns. from 1.3 ′ 10-7-1.3 ′ 10-5M for 3 h. A synergistic action was obsd. between II (1.3 ′ 10-6M) and zebrafish pituitary homogenate in stimulating GVBD. Moreover, pretreatment of the follicles with zebrafish pituitary homogenate enhanced the GVBD capacity of II. Apparently, one (or more) pituitary factor(s) stimulate(s) the synthesis of a deoxycorticosterone-like compd. or a metabolite which in turn initiates the maturation of oocytes.
