DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) Male and female rats received 50 mmol pentachloronitrobenzene [82-68-8]/kg by oral intubation daily for 7 days. The final excreta were hydrolyzed and analyzed by electron capture gas-liq. chromatog. for the presence of pentachlorobenzenethiol [133-49-3] and tetrachloro-1,4-benzenedithiol [67341-48-4] (derived from the equiv. N-acetylcysteine conjugates). No differences were found between the sexes for feces and bile but the urinary excretion of both thiols by females was >10-fold greater than males. A similar result for urine was obtained following i.p. administration of a single 20-mmol/kg dose of S-pentachlorophenyl-N-acetyl-L-cysteine (I) [68593-98-6]; in addn., cotreatment with probenecid did not greatly change excretion by the males but considerably reduced excretion by females. The sex difference in the urinary levels of I after 40 and 100 mmol/kg pentachloronitrobenzene was confirmed by high-performance liq. chromatog. of I and again probenecid inhibited the excretion. Anal. of urine by TLC following a dose of 14C-labeled hexachlorobenzene [118-74-1] (8 mCi/kg; 0.67 mmol/kg) showed that more radioactivity was assocd. with I from female rats than males. Thus, I, a metabolite of hexachlorobenzene and pentachloronitrobenzene, may be excreted by an active renal secretion which is particularly developed in female F344 rats.

DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) Female rats were dosed intraperitoneally with 14C-labeled hexachlorobenzene (I) [118-74-1]. The drug was administered on 2 or 3 occasions. The total doses amounted to 260 and 390 mg/kg. In urine pentachlorophenol [87-86-5], tetrachlorohydroquinone [87-87-6], andpentachlorothiophenol [133-49-3] were present as major metabolites. One of the isomers of tetrachlorothiophenol [60135-24-2] was present as a minor metabolite. In the feces pentachlorophenol and pentachlorothiophenol only were identified. Radioactive C excreted with urine and feces amounted to 7% and 27%, resp., of the radioactivity administered. More than 90% of C-14 excreted in urine was contained in the major metabolites. In the feces about 30% of the excreted radioactivity was bound to metabolites, and . 118-74-1 and 60135-24-2 which are cas registry numbers of chemicals are mentioned.apprx.70% was contained in the unchanged drug, while in the tissues of the animals only pentachlorophenol was detected in measurable amts., accounting for 10% of label in blood and <0.1% of C-14 detd. in body fat. Total radioactivity contained in the metabolites detected in the animal body and in the excreta accounted for .apprx.16% of the administered radioactivity. .
