DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) Bradykinin [58-82-2] stimulates [3H]thymidine incorporation and DNA synthesis in resting, serum-deprived NIL8 hamster cells. The ED50 for this stimulation is 4.52 nM. Other kinin peptides (including Lys-bradykinin (kallidin) [342-10-9] and Met-Lys-bradykinin [550-19-6]) also stimulate [3H]thymidine incorporation into the NIL8 cells, whereas desArg9-bradykinin [15958-92-6] is without effect, suggesting that the action of the kinin peptides is through type B2 receptors. Bradykinin also stimulates DNA synthesis in IMR-90 human fibroblasts; however, this effect is obsd. only in the presence of indomethacin, which blocks prostaglandin synthesis. Apparently, prostaglandins act as neg. modulators of the growth-stimulatory effects of bradykinin in the fibroblasts. This conclusion is supported by the observation that exogenously added PGE1 [745-65-3], PGE2 [363-24-6], PGA1 [14152-28-4], PGA2 [13345-50-1], PGB1 [13345-51-2], and PGB2 [13367-85-6] strongly inhibit [3H]thymidine incorporation in the human fibroblasts. The direct effect of bradykinin obsd. 745-65-3 and 58-82-2 are just another two chemicals used in this study. in the NIL8 cells may be attributable to the relative resistance of these cells to growth inhibition by prostaglandins. .
