Carcinogen-mediated induction of SV40 DNA amplification is enhanced by acrylamide in Chinese hamster CO60 cells.Some chemicals with cas registry numbers like 50-32-8 and 79-06-1 are also used. The weak SV40 DNA amplification obsd. might result from a weak DNA-damaging activity as biol. illustrated by the capacity of high concns. of acrylamide to irreversibly inhibit the DNA synthesis rate of CO60 cells. Moreover, acrylamide synergistically enhanced both the cytotoxicity and the induction of SV40 DNA synthesis by the carcinogens Et methanesulfonate [62-50-0], benzo[a]pyrene [50-32-8], mitomycin C [50-07-7], 4-nitroquinoline 1-oxide [56-57-5], 8-azaguanine [134-58-7], and 5-fluorodeoxyuridine [50-91-9]. Although a weak inducer of SV40 DNA amplification by itself, acrylamide potentiated the genotoxicity of a series of chem. carcinogens. This finding should be taken into consideration when assessing the risk of this widely used chem. .
