Deletions in the 5q21 region assocd. with colorectal cancer were found to map to a single polymorphic site found in a single cosmid. Subclones from this cosmid were used to detect changes in the region in carcinomas. By cross-hybridization of this clone with rat DNA and by sequencing of the cross-hybridizing regions a pair of exon-like open reading frames were found. PCR primers derived from these sequences were used to amplify rat mRNA and a transcript identified; the transcript was found in most tissues of the normal rat. A cDNA was cloned from human brain cDNA library in lZAP by screening with the PCR amplification product. Somatic variation was obsd. in the gene in colorectal cancers. .
