DOCUMENT TYPE: Conference CA Section: 3 (Biochemical Interactions) Neuronal response of the lateral geniculate nucleus of anesthetized cats to visual stimuli were investigated following iontophoretic application of glutamate [56-86-0], bicuculline [485-49-4], .gamma.-aminobutyrate [56-12-2], and (or) strychnine-HCl [1421-86-9] via micropipets positioned in the lateral geniculate body. The results suggest that .gamma.-aminobutyrate may be an inhibitory transmitter in this system, and support the concept that postsynaptic inhibition occurs in lateral geniculate nucleus cells. .

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Pretreatment with naloxone-HCl [357-08-4] at a subconvulsant dose (90 mg/kg, i.p., 5 min before challenge) reduced the ED50 for bicuculline [485-49-4] convulsions in mice, but had no effect on the dose-response for strychnine-HCl [1421-86-9]. In contrast, diazepam [439-14-5] pretreatment (5 mg/kg, i.p., 30 min before challenge) increased the ED50 of both bicuculline and naloxone, but had no effect on strychnine-induced convulsions. Bicuculline, naloxone, morphine [57-27-2], levorphanol [77-07-6], and dextrorphan [125-73-5] displaced 3H-labeled GABA [56-12-2] from GABA receptor sites in homogenates of human cerebellum, with 50% inhibition at 8, 308, 400, 250, and 300.mu.M resp. Addnl., microiontophoretically applied naloxone completely and reversibly antagonized GABA-evoked inhibition in neurons. The behavioral excitation obsd. after administration of large amts. of opiates or opiate antagonists may reflect functional blockade of GABA-inhibitory systems. .
