DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The blood levels and urinary excretion of the antimycobacterial drugs ethionamide [536-33-4] and prothionamide [14222-60-7] were compared after oral dosage in man. High-pressure liq. chromatog. methods were used to det. the 2 closely related thioamides and their microbiol. active sulfoxide metabolites after the ingestion of both single and combined doses of the 2 drugs. Both drugs were rapidly eliminated from the body, the half-life for the urinary excretion and removal from the plasma of prothionamide being slightly less than that of ethionamide. Less than 0.1% of the orally administered doses were excreted unchanged in the feces. Plasma concns. of ethionamide and its sulfoxide metabolite [536-28-7] were substantially higher than those of prothionamide and prothionamide sulfoxide [62178-61-4]. The implications of these findings for the use of ethionamide or prothionamide in the treatment of lepromatous leprosy are discussed.Except for chemicals metioned above, 536-33-4 and 536-28-7 are also used. .

DOCUMENT TYPE: Conference CA Section: 3 (Biochemical Interactions) Section cross-reference(s): 1 Isoprodian [55015-10-6] showed enhanced activity compared to that of its individual components (isoniazide [54-85-3], prothionamide [14222-60-7], and DDS [80-08-0]) against most Mycobacterium species tested. DDS appeared to be essential for this effect even though it alone showed almost no growth inhibitory activity against the organisms tested, with the exception of M. marinum. The combination of Isoprodian components appeared to increase the bacteriostatic effect rather than the killing rate. Some previous pharmacol. studies with Isoprodian in human volunteers are also reviewed.
