Five samples complied fully with the requirements, 2 samples failed, and 11 samples had minor shortcomings. This work is preceded by a discussion on the treatment of malaria and on the pharmacol., pharmacokinetics and metab., toxicol., and anal. of the title compds.

DOCUMENT TYPE: Journal CA Section: 63 (Pharmaceuticals) The biol. availability of quinidine [56-54-2] from a newly developed slow-release tablet (Optochinidin retard [50-54-4]) was compared with that from a std. slow-release quinidine speciality (SCR) in 20 healthy volunteers. The single oral dose administered was 1,000 mg quinidine. Both slow-release forms guaranteed a considerably delayed rise in the quinidine concns. in the plasma. On the av., .apprx.200 .mu.g quinidine/100 mL plasma was achieved only after 4 h. This delayed rise prevented too high, possibly cardiotoxic, tissue concns. After using a precise anal. procedure which detects only quinidine and dihydroquinidine [1435-55-8], the mean lower threshold was cor. to 150 .mu.g quinidine/100 mL plasma. After administration of SCR the quinidine level was above this threshold for a mean of 5 h, and after Optochinidin for more than 8 h. With a half-life of more than 11 h Optochinidine retard can be administered at 12 h intervals. The single initial dose should be twice the maintenance dose.
