DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) To assess possible risks involved in the administration of HI 6 (I) [34433-31-3] its degrdn. products were analyzed at pH 2.0, 4.0, 7.4, and 9.0. At pH 2.0, where HI 6 in aq. soln. has its maximal stability, attack on the aminal-acetal bond of the ether bridge predominates, with formation of HCHO [50-00-0], isonicotinamide [1453-82-3], and pyridine 2-aldoxime [873-69-8]. Besides, HI 6 decomps. at the oxime group yielding 2-cyanopyridine [100-70-9]. Liberation of hydrocyanic acid [74-90-8] at pH 2.0 is <5%. At pH 7.4, primary attack is on the oxime group, resulting in formation of the corresponding pyridone via an intermediate nitrile. The pyridone was isolated and identified as a 2-pyridinone deriv. [106915-64-4]. This major metabolite deaminates further to the 4'-carboxy-2-pyridinone deriv. [107089-73-6], which ultimately decomps. into isonicotinic acid [55-22-1], and 2-pyridone [142-08-5]. Hydrolysis of the acid amide group probably also occurs with HI 6 itself. Significant amts. of free hydrocyanic acid were only detected in the presence of an alkali trap; otherwise hydrocyanic acid reacts with HCHO to yield hydroxyacetonitrile [107-16-4] from which hydrocyanic acid can be liberated again. Up to 0.6 equiv of hydrocyanic acid were evolved at pH 7.4. After repetitive administration and impaired renal elimination of HI 6, e.g. during renal shock, there might be some risk of cyanide intoxication. .

DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) Relative stability studies of 3 organophosphate-inhibited acetylcholinesterase reactivators, 1-(2-hydroxyiminomethyl-1-pyridinium)-3-(4-carbamoyl-1-pyridinium) -2-oxapropane dichloride (HI 6)(I) [34433-31-3], 1,1'-trimethylenebis(4-hydroximinomethylpyridinium) dichloride (MMB 4) [51026-61-0], and 1,1'-trimethylenebis(4-hydroximinomethylpyridinium) dibromide (TMB 4) [56-97-3] were carried out by semiquant. TLC and NMR methods. TMB 4 appears to be the most, and HI 6 the least stable of the 3 compds. The extent of hydrolysis of HI 6, MMB 4, and TMB 4 in 0.05M, pH 7 phosphate buffer was ~50, 25, and <1%, resp., after 20 days at room temp. The hydrolysis products of HI 6 were identified by NMR spectrometry and mass spectroscopy (electron impact) as 2-pyridinealdoxime [873-69-8], picolinamide [1452-77-3], and isonicotinamide [1453-82-3], whereas that of MMB 4 was identified as 4-pyridinealdoxime [696-54-8]. The stability of these reactivators decreases with increasing pH. TMB 4 was stable under both neutral and basic conditions at room temp. Deuterium exchange of the methylene protons of MMB 4 in D2O and of the protons at the 2- and 6-positions of the pyridinium ring of TMB 4 in NaOD/D2O were obsd. .
