DOCUMENT TYPE: Journal CA Section: 3 (Biochemical Genetics) Section cross-reference(s): 13 Two different drugs [b-aspartylhydroxamate (b-AHA) [1955-68-6] and albizziin (Alb) [1483-07-4]] and 2 selective protocols were used to select mutations involving asparagine synthetase (AS) [9023-69-2] in CHO cells. With b-AHA, single-step mutants were produced that were resistant to the drug due to structural changes in AS. Mutant selection with Alb gave resistant lines with elevated AS activities and large, homogeneously staining regions in the karyotype. These mutants led to the development of 2 methods for cloning the AS gene. One method involves rescuing the gene (by selecting for b-AHA resistance) after transfer into COS-1 cells. The other method involves constructing cDNA clones from mRNA isolated from the highly amplified Alb-resistant lines on the assumption that the 300-fold increase in enzyme activity is reflected in a comparable increase in mRNA.

I was active against vaccinia and influenza A viral infections and II against Newcastle disease, vaccinia, influenza A, herpes, and infectious pancreatic necrosis viral infections. L-b-Ureidoalanine [1483-07-4] showed activity against Newcastle disease and herpes viral infections.
