Thus, II 1816, Na2CO3 606.3, and DMF 11,470 parts were heated to 75-80°, bubbled with N for ~30 h, cooled, mixed with 16,100 parts H2O, and cooled to give 97% cryst. I. Cl2CO was bubbled at 0.52-1.63 parts/h through a mixt. of I 1, H2O 2.9-6.3, CH2Cl2 4.7-9, PhOH 0.008, Et3N 0.005-0.008, and Na gluconate 0.002 part to give a polycarbonate (III) [31546-39-1] of intrinsic viscosity ~0.52 dL/g (CH2Cl2, 25°). III was extruded and pelletized and the pellets were molded into test specimens having Izod notched impact strength 15 ft-lb./in.

DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) The role of metab. in the estrogenic activity of chlorinated hydrocarbon pesticides was examd. Whether the estrogenic activity in tech. grade prepns. of the pesticide methoxychlor (I) [72-43-5] is due to I or to contaminants was also investigated. Identified compds. in tech. I were examd. by an in vitro method to det. whether they are estrogens or proestrogens. This method showed that purified I and 1,1-dichloro-2,2-bis(4-methoxyphenyl)ethene (MDDE) [2132-70-9], are proestrogens and that monohydroxymethoxychlor [28463-03-8] and monohydroxy-MDDE [75938-34-0] are estrogens. Thus, the estrogenic activity in tech. I is due to both I and contaminants. MDDE is an in vivo metabolite of I, and the mono- and bishydroxy [1,1,1-trichloro-2,2-bis(4-hydroxyphenyl)ethane [2971-36-0]] derivs. of I and MDDE are metabolites of I and MDDE, resp. These metabolites exhibited in vitro estrogenic activity in the following order of potency: bishydroxy-MDDE [14868-03-2] > bis-hydroxy I > mono-hydroxy-MDDE > mono-hydroxy I. A similar order of potency was obsd. in vivo, demonstrating that metabolites of I are potent estrogens. In addn. to phenolic products, hepatic monooxygenase [9038-14-6] metabolize I and MDDE to reactive intermediates that bind covalently to microsomal proteins. Further studies are needed to det. the factors controlling the 2 pathways of I metab. and det. whether covalent binding is assocd. with cellular and organ toxicity. .
