Twelve days following application of either prepn. the total no. of ribosomes was increased 30% in comparison with controls. The RNA precursors orotic acid [65-86-1], Na orotate [154-85-8], and methylglucamine orotate [59404-17-0] also stimulated differentiation and maturation of neurons but the effect differed somewhat from that of protein synthesis activators, esp. on ribosomes.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) In order to obtain further evidence of the pharmacol. effects of retention-facilitating substances, the influence of Na orotate [154-85-8], methylglucamine orotate [59404-17-0], and pirazetam (I) [7491-74-9] on the bioelec. activity of central nervous system structures were studied. Na orotate showed no influence on the cortical and hippocampal spontaneous electroencephalog. of conscious nonimmobilized rabbits. The amplitude of the 1st surface-pos. wave of the cortical evoked potential that was elicited by stimulation of tooth pulp was, however, significantly increased in long-lasting terms by both I and the 2 salts of orotic acid. Similarly, the 3 substances also influenced the changes in excitability of tooth pulp. Thus, these chems. facilitate in a nonsp. manner responses to externally applied stimuli through central structures. The relations between the altered stimulus response and the retention-facilitating action of these substances are discussed.
