Cellular interactions of auranofin and a related gold complex with RAW 264
Cellular interactions of auranofin and a related gold complex with RAW 264.7 macrophages. Snyder, Rosanne M.; Mirabelli, Christopher K.; Crooke, Stanley T. (Dep. Mol. Pharmacol., Smith Kline and French Lab., Philadelphia, PA 19101, USA). Biochem. Pharmacol., 36(5), 647-54 (English) 1987. CODEN: BCPCA6. ISSN: 0006-2952. DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The effects of auranofin (AF) [34031-32-8] and triethylphosphine gold chloride (TEPG) [15529-90-5] on macrophage-like RAW 264.7 cells were studied. More gold from TEPG than AF assocd. with cells over time and at a variety of concns. The cellular assocn. of AF and TEPG was temp. dependent; the energy of activation for cell assocn., the rate-limiting step in the thiol exchange process, was lower for TEPG than AF. Cellular assocn. and uptake of both compds. did not require metabolic energy. Efflux of both AF and TEPG was time-, temp.In this experiment, several chemicals are used like 70-18-8 and 7440-57-5 -, and thiol-dependent. Sequential thiol exchange may be a generic phenomenon for cellular uptake and distribution of thiol reactive gold compds. and the rate-limiting step is the exchange of either tetraacetylthioglucose [19879-84-6] or chloride for a membrane-localized sulfhydryl group. .
The effect of drugs in established rabbit monoarticular arthritis
The effect of drugs in established rabbit monoarticular arthritis. Blackham, Alan; Radziwonik, Henryk (Dep. Biochem. Pharmacol., Fisons Ltd., Loughborough/Leicestershire, Engl.). Agents Actions, 7(4), 473-80 (English) 1977. CODEN: AGACBH. DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Established antigen-induced arthritis was produced in 264 out of 339 sensitized rabbits 4-6 wk after a single intraarticular injection of ovalbumin into 1 knee joint. Pos. arthritis was diagnosed when the joint swelling at this time was 32 mm. A pos. correlation between established joint swelling and delayed hypersensitivity to ovalbumin injected intradermally 24 h prior to joint challenge was demonstrated. Groups of 5-10 arthritic rabbits were dosed orally with increasing or fixed doses of a range of drugs for a period of 3-7 wk. Drug activity was measured on joint swelling intermittently during the test and on joint macroscopy, cell content of the synovial fluid and joint histopathol. at the end of the expt. Prednisolone sodium phosphate [125-02-0] and ketoprofen [22071-15-4] were the most active compds. tested. Chloroquine diphosphate [50-63-5], cyclophosphamide [50-18-0], indomethacin [53-86-1], naproxen [22204-53-1], SKF 36914 [15529-90-5], and sudoxicam [34042-85-8] had some activity, while aspirin [50-78-2], levamisole [14769-73-4], oxisuran [27302-90-5] and D-(-)-penicillamine-HCl [2219-30-9] had little or no activity.
