DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Ryanodine [15662-33-6] inhibited both release and uptake of Ca2+ by the sarcoplasmic reticulum in isolated cat papillary muscle. Elevation of high-energy phosphate secondary to decreased energy requirements was due to decreased Ca2+ availability to the myofilaments during systole. The slowed or incomplete relaxation caused by ryanodine may be a stimulus for myosin phosphorylation in isolated rat heart. Ryanodine probably decreases Ca2+ movement through the sarcolemma and so increases adenosine and inorg. P and decreases cAMP concn. in the myocardium. The effect of ryanodine on altered loading conditions and contraction velocities can be understood in terms of decreased Ca2+ availability to the myofilaments.

These data suggest that the antagonism is mediated by ryanodine-induced alterations in Ca2+ release from the sarcoplasmic reticulum.
