DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Section cross-reference(s): 4 The mutagenicity of pyrazole (I) [288-13-1] and 7 I derivs. (4-nitropyrazole [2075-46-9], 4-bromopyrazole [2075-45-8], 1-methyl-4-nitropyrazole [3994-50-1], 3,5-dimethyl-4-nitropyrazole [14531-55-6], 1-methyl-4-bromopyrazole [15803-02-8], 4,4'-dinitro-1,1'-methylenedipyrazole [86111-62-8], and 4,4'-dibromo-1,1'-methylenedipyrazole [86111-61-7]) was investigated with the L-arabinose forward mutation assay of S. typhimurium. Two nitroimadazoles (1-methyl-5-nitroimidazole [3034-42-2] and metronidazole [443-48-1]) were included as ref. drugs. The mutagenicity of each chem. was detd. by both preincubation and liq. tests, in the presence or absence of S9 microsomal fraction. The mutagenic response was expressed as the abs. no. of L-arabinose-resistant mutants growing in selective plates, supplemented with traces of D-glucose. Strain BA13 with a wild-type lipopolysaccharide barrier was used as a comparison to the deep rough deriv. BA9. No mutagenic effect was detected with I and 2 of its derivs., 1-methyl-4-bromopyrazole and 4,4'-dibromo-1,1'-methylenedipyrazole. The other 5 I derivs. were mutagenic to different degrees, although their mutagenic potencies were always considerably lower than those of the 2 nitroimidazoles. 4-Nitropyrazoles, as well as 4,4'-dinitro-1,1'-methylenedipyrazoles are promising as alternatives to, or even substitutes for, the currently used nitroimidazoles. .
